Use of cyclodextrins in ITP analysis

Abstract:

Tato bakalářská práce se věnuje možnostem použití cyklodextrinů v izotachoforetické analýze s vodivostní detekcí. Cílem bylo testování cyklodextrinů pro analýzu aminoderivátů adamantanu, amantadinu a rimantadinu, a následný vývoj a optimalizace metody pro separaci a stanovení těchto derivátů. V teoretické části je zpracovaná rešerše, v níž je věnována pozornost principu elektromigrační separační metody izotachoforézy a její instrumentaci. Dále jsou zde shrnuty poznatky o historii, vlastnostech a velmi širokém vyžití cyklodextrinů. Závěrem teoretické části jsou popsány vlastnosti amantadinu a rimantadinu, možnost jejich použití k léčbě chřipky a analytické metody používané pro jejich analýzu. Experimentální část se zabývá stanovením amantadinu a rimantadinu jednotlivě a následně také ve směsi. V obou případech byla provedena měření bez použití cyklodextrinů a s přídavkem β-cyklodextrinu do elektrolytového systému.

Abstract:

This bachelor thesis deals with possibilities of the use of cyclodextrins in isotachophoretic analysis with conductivity detection. The goal was testing of cyclodextrins for the analysis of amino derivatives of adamantane, amantadine and rimantadine, and subsequent development and optimization of a method for separation and determination of these derivatives. In the theoretical part is literature review in which attention is paid to the principle of electromigration separation method isotachophoresis and its instrumentation. Then there is a summary of the knowledge of the history, characteristics and very wide use of cyclodextrins. In the end of the theoretical part are described properties of amantadine and rimantadine, possibility of their use in the treatment of influenza and analytical methods used for their analysis. Experimental part deals with the determination of amantadine and rimantadine separately as well as in the mixture. Measurements were done both with and without the addition of cyclodextrins to the electrolyte system.
