Production of LL-37 and verification of its antimicrobial activity in vitro.

Abstract:

Možné řešení celosvětového problému bakteriální rezistence vůči antibiotikům nabízejí antimikrobiální peptidy, včetně LL-37, kterým se zabývá tato práce. Cílem práce je poskytnout přehled o antimikrobiálních peptidech, především o LL-37, jako možné alternativě antibiotik. Dále provést produkci LL-37 v Escherichia coli pomocí rekombinantních technologií a závěrem změřit antimikrobiální aktivitu peptidu proti vybraným patogenům. Teoretická část práce se zabývá historií léčby infekčních onemocnění, historií antibiotik, alternativními antimikrobiálními terapiemi, do kterých patří popis a analýza antimikrobiálních peptidů včetně LL-37. V praktické části je produkován LL-37 za použití metod genetického inženýrství. Následně je peptid LL-37 uvolňován od fúzního proteinu pomocí chemického štěpení, a nakonec je antimikrobiální aktivita měřena metodou redukce zákalu. Způsob produkce peptidu LL-37 použitý v této práci umožňuje snížit finanční náklady na jeho výrobu ve srovnání s jeho chemickou syntézou. V neposlední řadě, snížení nákladů na výrobu může přiblížit použití peptidu v klinické aplikaci.

Abstract:

The present thesis deals with antimicrobial peptides, including LL-37, which represent one of possible solutions to the worldwide problem of bacterial resistance to antibiotics. The aim of the thesis is threefold: firstly, to provide an overview of antimicrobial peptides, especially LL-37, as a possible alternative to antibiotics; secondly, to produce LL-37 in Escherichia coli using recombinant technologies; and thirdly, to measure the antimicrobial activity of the peptide against selected pathogens. The theoretical part deals with the history of treatment of infectious diseases, the history of antibiotics, alternative antimicrobial therapies, which include the description and analysis of antimicrobial peptides, including LL-37. In the practical part, LL-37 is produced using genetic engineering methods. Then, the LL-37 peptide is released from the fusion protein by chemical cleavage, and finally, the antimicrobial activity is measured by a turbidity assay. The production method of the LL-37 peptide discussed in the thesis leads to the reduction of financial costs of its production in comparison with its chemical synthesis. Last but not the least, the reduction of manufacturing costs may also lead to the use of the peptide in clinical application.
