Genotyping of genus Borrelia in Czech republic territory

Abstract:

Spirocheta Borrelia burgdorferi je nejčastějším původcem Lymeské boreliózy. Lymeská borelióza je multisystémové onemocnění a je v současné době nejrozšířenější zoonózou v Evropě a Spojených státech. Na člověka je borélie přenášena hematofágními členovci, převážně klíšťaty rodu Ixodes. Dalšími hostiteli borélií jsou savci jako je vysoká zvěř, psi a kočky, ptáci a hlodavci. Z klíšťat zasílaných lidmi z celé České republiky bylo izolováno 89 vzorků DNA borélií. Genotypizace těchto vzorků byla provedena metodou Real-time PCR, amplifikací IGS částí DNA borélií, a následnou sekvenační analýzou těchto amplifikátů. S 56 získanými sekvencemi byl vytvořen fylogenetický strom rodu Borrelia na území České republiky. Z 56 vzorků DNA borélií bylo 49 vzorků identifikováno jako B. afzelii, 4 vzorky jako B. garinii, 2 vzorky jako B. valaisiana a po 1 vzorku jako B. spielmanii, B. duttonii, B. lusitaniae a blíže nespecifikovaný druh B. burgdorferi. Z výsledků vyplývá, že na území České republiky je nejvíce rozšířená borélie je B. afzelii.

Abstract:

Spirochetes Borrelia burgdorferi are the caustative agents of Lyme disease. Lyme disease, a multisystemic disorder, is the most prevalent vector-borne disease of humans in the United States and Europe. Into humans, Borrelia is most often transmitted by hematofagous arthropods, especially ticks of the Ixodes genus. Other Borrelia hosts are mammals such as deer, dogs and cats, birds, and rodents. 89 samples of borrelian DNA were izolated from tikcs that have been sent to the laboratory by people from the territory of Czech republic. Genotyping of these borrelian DNA samples was performed using the Real-time PCR, targeting the IGS regions, and the sequence analysis of these particular DNA regions. Out of 89 sequences of the borrelian DNA 56 sequences were used to create the phylogenetic tree of the Borrelia genus in the territory of Czech republic. Out of 56 samples of the borrelian DNA 49 samples were identified as B. afzelii, 4 samples as B. garinii, 2 samples as B. valaisiana and 1 sample of each as B. spielmanii, B. duttonii, B. lusitaniae, and not-specified B. burgdorferi type. The results shows that the most frequently occurring type of the Borrelia genus in the Czech republic was B. afzelii.
