Cloning and expression of glycosyltransferases

Abstract:

Oligosacharidy mají v lidském těle důležitou biologickou úlohu. Mají jednak funkci strukturní a zásobní, ale také funkci signalizační, komunikační, rozpoznávací a další. Jsou důležité v imunitním systému, morfogenezi nebo také třeba v kancerogenezi a jsou také součástí struktur antigenů různých systémů krevních skupin. Glykosyltransferasy jsou enzymy, které jsou zodpovědné za syntézu těchto oligosacharidů, ale také polysacharidů a glykokonjugátů. Tyto enzymy katalyzují proces přenosu cukerných zbytků z aktivovaného nukleotid-cukru na akceptor, což může být rostoucí oligosacharid, lipid nebo bílkovina. Protože se glykosyltransferasy nacházejí převážně na membránách a jsou hydrofobní, je jejich příprava a práce s nimi do jisté míry obtížná. Při expresi v hostitelských buňkách totiž dochází často k tvorbě inkluzních tělísek. Diplomová práce byla zaměřena na přípravu eukaryotní rekombinantní xylosyltransferasy z Caenorhabditis elegans, což je červ patřící do kmene hlístů, který je využíván jako modelový organismus pro vyšší živočichy. Jako hostitelský organismus byly využívány buňky E coli. Získáním čisté xylosyltransferasy by byla možná její charakterizace, jak strukturní tak i funkční.

Abstract:

Glycosyltransferases are the enzymes that are involved in the biosynthesis of oligosaccharides, polysaccharides and glycoconjugates. These enzymes of enormous diversity mediate a lot of functions from structure and storage to recognition and specific signalling. Glycosyltransferases transfer the sugar moiety from an activated nukleotide-sugar to an acceptor, which can be a growing oligosaccharide, a lipid or a protein. GTs are present in both prokaryotes and eukaryotes. In eukaryotes, most of the glycosylation reactions that generate the diversity of oligosaccharide structures of eukaryotic cells occur in the Golgi apparatus. Since GTs are generally transmembrane proteins and therefore hydrophobic, their expression in host organisms is very problematic for thein toxicity and forming of inclusion bodies. The thesis was focused on the preparation of eukaryotic recombinant xylosyltransferase from Caenorhabditis elegans, which serves as a model genetic organism for higher animals. E. coli was used as a host organism. XylT was expressed in ArcticExpressTM (DE3)RIL Competent Cells a ArticExpressTM (DE3) Competent Cells. The expression proceeded at the temperature of 10 – 13 C, because low-temperature cultivation represents one strategy for increasing the recovery of soluble protein. Part of the protein was expressed into inclusion bodies. Therefore, we made both native purification from cytosol and refolding on column from inclusion bodies. Because of low purity, it was necessary to use another method for separation from other proteins. There was used a gel permeation chromatography, but purification didn't lead to the pure protein. For future, there are needed other purifying steps.
