Karyotype evolution and interphase chromosome organization in plants

Abstract:

U Brassicaceae byl u Arabidopsis thaliana prokázán chromocentrický (CC) smyčkový model nebo konfigurace podobná růžici. Tento vzorec je charakterizován telomerami shlukujícími se kolem jádra, zatímco centromery jsou umístěny na periferii jádra - centromerický heterochromatin tvoří chromocentrum. Euchromatinový prstenec o délce 0,2 až 2 Mb vychází z chromocentra a tvoří strukturu podobnou růžici. Až dosud nevíme, zda je organizace podobná růžici v cruciferu univerzální. V této diplomové práci se snažíme tuto mezeru vyplnit pomocí fluorescenční hybridizace in situ u sedmi diploidních druhů (2n = 10-16) u Brassicaceae. Tyto druhy představují hlavní kmeny křižáků s různými velikostmi genomu (160 Mb - 4 260 Mb) a fylogenetickými polohami. Výsledky ukázaly, že shlukování telomer spojených s nukleolem bylo potvrzeno u arabidopsis (157 Mb) a bylo nově identifikováno jako hlavní jaderný fenotyp u jiných druhů s malým genomem (215-381 Mb) tří linií (I, III a IV). U druhů velkého genomu (2 611-4 264 Mb) chybělo shlukování telomer spojené s nukleolem a přijaly buď konfiguraci podobnou Rabl, nebo rozptýlenou distribuci v nitru jádra. U Arabis cypria (381 Mb) a Bunias orientalis (2 611 Mb) byly v prašníkových a kmenových tkáních pozorovány tkáňově specifické vzory odchylující se od hlavních jaderných fenotypů. Data prezentovaná v práci celkově charakterizují, že mezifázovou jadernou organizaci určuje konfigurace podobná růžici, včetně shlukování telomérů spojených s nukleolem v druzích malého genomu z různých intrafamiliálních kladů, a identifikace těchto genomových vlastností spíše než fylogenetické polohy.

Abstract:

In Brassicaceae, chromocenter (CC)-loop model or rosette-like configuration was proved in Arabidopsis thaliana. This pattern is characterized by telomeres clustering around the nucleolus, while centromeres are located at the periphery of the nucleus—centromeric heterochromatin forms chromocenter. The euchromatin ring with a length of 0.2 to 2 Mb emanates from the chromocenter, forming a rosette-like structure. Until now, we do not know whether the rosette-like organization is universal in crucifer. In this thesis, we aim to fill this gap through the use of fluorescence in situ hybridization in seven diploid species (2n = 10 - 16) in Brassicaceae. The species represent major crucifer clades with different genome sizes (160 Mb – 4 260 Mb) and phylogenetic positions. The results showed that nucleolus-associated telomere clustering was confirmed in arabidopsis (157 Mb) and was newly identified as the major nuclear phenotype in other species with a small genome (215 – 381 Mb) of three lineages (I, III, and IV). In large genome species (2 611 – 4 264 Mb), nucleolus-associated telomere clustering was absent, and they adopted either a Rabl-like configuration or dispersed distribution in the nuclear interior. In Arabis cypria (381 Mb) and Bunias orientalis (2 611 Mb), tissue-specific patterns deviating from the major nuclear phenotypes were observed in anther and stem tissues, respectively. Altogether the data presented in the thesis characterize that rosette-like configuration, including nucleolus-associated telomere clustering in small-genome species from different intrafamilial clades, and identify those genomic properties rather than phylogenetic position, determine the interphase nuclear organization.
