Potential utilization of insects and their symbionts in biofuel production

Abstract:

Fosilní paliva nejsou nevyčerpatelná a je tedy nutné hledat alternativy, které by je mohly nahradit. Jako jedna z?možností se nabízí transesterifikace tukového tělesa hmyzu. V?efektivitě tohoto procesu by mohli hrát roli střevní symbionti hmyzu, kteří napomáhají trávení a zlepšují tak metabolismus, a tedy i růst tukového tělesa. Prvním cílem práce bylo zpracování rešerše týkající se využití hmyzu v?procesu výroby biopaliv a následně i symbiontů, kteří by v?tomto procesu mohli být užiteční. Druhým cílem byl návrh experimentu, jehož součástí je výběr vhodného druhu hmyzu, dále odchov larev, výběr vhodných symbiontů a následně proces extrakce tukového tělesa a výroba biopaliv. Dle dostupné literatury se nejvhodnějšími kandidáty zdají být larvy Hermetia illucens a Tenebrio molitor, přičemž larvy H. illucens byly následně zařazeny do samotného návrhu experimentu. Jako vhodní symbionti byli do experimentu vybráni Bacillus subtilis a Rhodococcus rhodochrous. Nejvhodnější metodou se zdá být metoda Soxhletova, která je vhodnější pro menší množství larev. Využitím kombinace těchto parametrů by mohlo být možné docílit lepší biokonverze přijaté potravy, lepšího růstu tukového tělesa a kvalitnější extrakce tohoto tuku. Toto by vedlo k?zisku většího množství paliva. Využití tukového tělesa je stále poměrně nový koncept a je zde mnoho možností pro další výzkum ať už v?rámci hostitelů, možných symbiontů či alternativních způsobů extrakce tuku.

Abstract:

Fossil fuels are not inexhaustible, and it is necessary to search for alternatives. One of the alternatives is the transesterification of insect fat body. Insect gut symbionts could play a role in the effectiveness of this process, which could help with digestion and improve metabolism, therefore even the growth of the fat body. The first goal of the thesis was to create a review regarding the usage of insects in the process of biofuel production and subsequently on symbionts that could be helpful in this process. The second goal was to design the experiment, which included a selection of suitable insect species, then larvae rearing, selection of suitable symbionts and subsequently process of fat body extraction and fuel production. According to available literature, the best candidates are larvae of Hermetia illucens and Tenebrio molitor, whereby larvae of H. illucens were included in the design of the experiment. As suitable symbionts in the experiment were chosen Bacillus subtilis and Rhodococcus rhodochrous. The most suitable method seems to be the Soxhlet method, which is better for smaller amounts of larvae. With the usage of a combination of these parameters may be possible to gain better bioconversion of ingested food, better growth of fat body and better quality of fat extraction. This could lead to better fuel gain. Usage of the fat body is still a proportionately new concept and there are a lot of opportunities for next research whether within hosts and possible symbionts or alternative methods of fat extraction.
