Utilization of molecular genetic markers in the process of improoving the quality production of the selected brassica vegetable seeds

Abstract:

Obsahem této disertační práce bylo navrhnout využití molekulárně genetických markerů SSR (Simple Sequence Repeats) pro identifikaci rodičovských linií a hybridních jedinců odrůd zelí hlávkového. Dále byly testovány způsoby detekce a možnosti eliminace bateriálního patogenu Xantomonas campestris pv. campestris (Xcc) z osiva brukvovitých zelenin. V první části literární rešerše je uveden přehled systémů využívajících molekulární markery při určování genetické čistoty rostlinného materiálu. Druhá část se zabývá problematikou Xcc v souvislosti s šířením této bakterie prostřednictvím osiva a rozmnožovacího materiálu. Jsou zde uvedeny nástroje pro determinaci bakteriálních patogenů rostlin z čeledi Brassicaceae s důrazem na využití molekulární genetiky a přehled postupů pro identifikaci Xcc v osivu brukvovitých zelenin. Ve třetí části jsou uvedeny metody pro kontrolu Xcc v osivu zahrnující fyzikální a biologické postupy a také možnosti ošetření rostlinného materiálu s využitím přípravků obsahujících nanočástice. V experimentální části byly optimalizovány postupy pro PCR analýzu mikrosatelitní DNA klíčních rostlin zelí hlávkového. Temlátová DNA byla získána optimalizací protokolu pro expresní extrakci nukleových kyselin. Pro detekci a identifikaci Xcc v rostlinném materiálu byly využity postupy zakotvené v oficiálních protokolech ISTA (International Seed Testing Association) zahnující kultivační techniky, a molekulární analýzy PCR a Real-time PCR. Za účelem otestovat efekt vůči Xcc bylo uměle inokulované osivo ošetřeno horkou vodou, vysokými tlaky, ozonem nebo nanočásticemi. Jako dílčí výsledek této práce byl vypracován postup pro rozlišení celkem 24 genotypů zelí hlávkového představující 8 hybridních odrůd a jejich rodičovské linie s využitím SSR markerů. Tímto postupem byla otestována hybridnost celkem u 20 šarží osiva dodaného produkční společností. V rámci monitoringu Xcc byl proveden epidemiologický průzkum na produkčních plochách brukrovité zeleniny na území ČR. Systémy pro detekci a identifikaci bakterie Xcc byly rovněž aplikovány celkem u 42 šarží osiva brukvovitých zelenin. Z hlediska účinnosti eliminace Xcc v osivu byla jako nejvhodnější způsob ošetření doporučena metoda aplikace stříbrných nanočástic, pomocí které bylo inokulum Xcc nejvíce redukováno a oproti ostatním metodám zde nebyl pozorován negativní dopad na klíčivost osiva.

Abstract:

The aim of this thesis was to design the system based on molecular genetic markers SSR (Simple Sequence Repeats) for the identification of parental lines and hybrids of the cabbage. Subsequently, the possibilities of detection and elimination of bacterial pathogen Xanthomonas campestris pv. campestris (Xcc) in cruciferous seeds were tested. First part of the literary research summarize the systems of molecular markers used for the determination of plant material genetic purity. Second part deals with topic of spreading the Xcc through the seeds and propagation material. The tools for the determination of bacterial pathogens of the Brassicaceae family were summarized and the importance of the molecular genetics for the Xcc identification was highlighted. In the third part the methods for the control of the Xcc in the plant material including physical, biological and nanoparticle approaches were described. For the experimental part, the optimization of the PCR analysis of microsatellite DNA of the cabbage seedlings was carried out. Template DNA was obtained using the optimized prototocol for the express extraction of the nucleid acids. For the detection and identification of Xcc in the plant material the official approaches of ISTA (International Seed Tested Association) including cultivation techniques, PCR and Real-time PCR were used. For the testing of effectiveness against the Xcc the artificially inoculated seeds were treated by hot water, high pressure, ozone or nanoparticles. Partial results constitute of development of the SSR marker-based method for the distinguishing of 24 cabbage genotypes representing 8 hybrid varieties and their parental lines. The hybridity of 20 seed batches supplied by the commercial company were tested by this method. Within the monitoring of the Xcc occurrence the epidemiological research in the production fields of the Czech Republic was carried out. The Xcc detection systems were also applied on 42 cruciferous seed batches. For the elimination of Xcc inoculum from the seeds the application of the silver nanoparticles was evaluated as the most effective one. In comparison to other treatments the application of the silver nanoparticles didn’t influence the seed germination.
