Glioma Cancer Stem Cells – Their Characteristic, Analysis and Isolation

Abstract:

Tato diplomová práce se věnuje nádorovým kmenovým buňkám gliomů u myší gliomové linie GL261. Cílem této práce je na základě specifických povrchových markerů charakterizovat tyto nádorové kmenové buňky, následně je pomocí těchto markerů izolovat a nakonec je analyzovat pomocí exprese ABC transportérů a molekul specifických pro kmenové buňky. Na základě tohoto výzkumu bylo zjištěno, že v buněčné linii GL261 se nacházejí buňky, které na svém povrchu exprimují molekuly CD15, CD133 a A2B5, specifické pro neurální kmenové a progenitorové buňky a ABCG2 transportér. Některé z těchto buněk pozitivních na expresi daných povrchových molekul byly schopny vytvářet plovoucí sférické útvary zvané neurosféry. U vzniklých neurosfér byla zjištěna zvýšená exprese některých markerů pluripotence a markerů neurálních kmenových a progenitorových buněk, avšak byla zjištěna také zvýšená exprese některých molekul specifických pro diferencované neurální buňky. Z těchto výsledků tedy vyplývá, že v myší gliomové linii GL261 jsou přítomny tumorigenní buňky s charakteristikou nádorových kmenových buněk.

Abstract:

This thesis deals with cancer stem cells in murine glioma cell line GL261. The goal of this thesis is to characterise these cancer stem cells based on their specific surface markers, then isolate these cells via these markers and finally analyze them, especially through expression of ABC transporters and specific stem cell molecules. This research revealed, that within GL261 cell line, there are some cells expressing surface markers CD15, CD133, and A2B5, which are specific for neural stem and progenitor cells. Expression of the ABCG2 transporter was also detected. Some of the cells expressing these molecules were capable of producing floating spherical units, the so called neurospheres. In neurospeheres, the higher expression of some pluripotency markers and neural stem and progenitor cells markers was observed, however, also higher expression of some molecules specific for differentiated nerual cells was detected. Based on these findings, it is evident, that there are existing tumorigenic cells with the characteristic of cancer stem cells within murine glioma GL261 cell line.
