Hodnocení efektů cyanotoxinů na autotrofní organismy pomocí biochemických parametrů

Abstract:

Abstrakt Sinice jsou organismy produkující široké spektrum biologicky aktivních látek, kterými mohou ovlivňovat své okolí. Tyto sloučeniny mohou být výhodou v kompetičním boji proti konkurentům. Látky, které ovlivňují růst nebo vývoj jiného fytoplanktonního druhu, označujeme jako sloučeniny alelopatického významu. Alelopatický efekt vykazují nejen látky strukturně identifikované, ale také extracelulární produkty (exudáty) nebo extrakty různých druhů sinic. V rámci této diplomové práce byl studován vliv microcystinu na fytoplanktonní druhy. Testovali jsme vliv směsi microcystinů, jednotlivých strukturních variant microcystinu (MC-LR, MC-RR), exudátů různých druhů sinic a surového extraktu cyanobaktérie Microcystis aeruginosa na vybrané laboratorně kultivované zástupce fytoplanktonu (Chlorophyta, Cyanobacteria). Experimentální část diplomové práce můžeme rozdělit do několika celků, ve kterých jsme sledovali různě úrovně působení microcystinů - růst, biochemické parametry a diferenciaci buněk vláknitých sinic. V pokusu se směsí microcystinů a jeho purifikovaných strukturních variant MC-LR, MC-RR zaměřeném na ovlivnění růstu 6 zástupců fytoplanktonu byl pozorován inhibiční účinek zejména v přírodně nerelevantních koncentracích (25000 ug/l) od 7.dne expozice. Nejvíce inhibičně působil MC-RR, směs microcystinů inhibovala růst v menší míře. MC-LR prokazatelně ovlivnil pouze citlivější organismy. Při testování exudátů vybraných sinic Cylindrospermopsis, Microcystis, Aphanizomenon byl pozorován stimulační účinek na růst řas. Tento efekt byl koncentračně závislý u všech typů exudátů na všech testovaných kulturách. Při sledování změn vybraných detoxifikačních a antioxidačníchparametrů (glutathion, glutathion-S-transferáza a glutathion peroxidáza) vlivem směsi microcystinů, exudátu a surového extraktu Microcystis aeruginosa (konc. MC 300 ug/l), modifikovaly všechny testované varianty hladiny a aktivity sledovaných parametrů. U kultury vláknité sinice Trichormus variabilis jsme pozorovali u variant s microcystinem sníženou tvorbu akinet oproti kontrole. Tvorba heterocytů nebyla microcystinem ovlivněna. V tomto experimentu byly použity přírodně relevantní koncentrace microcystinu (0,2-20 ug/l).

Abstract:

Cyanobacteria are organisms which produce a wide spectrum of biologically active compounds. These compounds could influence the surroundings of cyanobacteria. They could also be an advantage in competition among organisms. Compounds which could affect growth or development of another phytoplankton species are called compounds with allelopatic properties. Allelopatic effects can be caused by compounds with identified structure and also extracellular products (exudates) or extracts of various cyanobacteria species. In the scope of this diploma theses the effect of microcystin at phytoplankton species was studied. We have tested effects of the microcystin mixture, its individual structural variants (MC-LR, MC-RR), exudates of various cyanobacteria species and crude extract of cyanobacteria Microcystis aeruginosa on selected laboratory cultivated phytoplankton representatives (Chlorophyta and Cyanobacteria). Experimental part of this study could be divided into several sections, in which the different levels of microcystin effects were studied (modification of growth, biochemical parameters and differentiation of filamented cyanobacteria). The experiment with microcystin mixture and its purified structural variants MC-LR, MC-RR was concerned with their effects on growth of 6 phytoplankton representatives. In this experiment inhibition effect mainly occurred just in cases of environmentally not relevant concentrations (25000 ug/l) from the seventh exposure day. The highest inhibition effect was observed after exposure to microcystin MC-RR. The mixture of microcystins caused less growth inhibitions MC-LR affected just the more sensitive organisms. Stimulation effect on algae growth was observed after exposure to the exudates from selected cyanobacteria Cylindrospermopsis, Microcystis and Aphanizomenon. This effect was concentration dependent for all tested exudates and all tested organisms. The study comparing selected detoxification and antioxidative parameters (glutathione, glutathione-S-transferase and glutathione peroxidase) after exposure to microcystin mixture, exudate and crude extract of Microcystis aeruginosa (conc. MC 300 ug/l) showed significant modification of levels and activities of tested parameters for all exposure variants. The effect of environmentally relevant concentrations of microcystin (0,2-20 ug/l) on cell differentiation of filamentous cyanobacteria Trichormus variabilis was tested. We observed decreased differentiation to akinetes after treatment with microcystin compared to control. The heterocyst frequency was not affected by microcystin.
