Abstract:

CRISPR/Cas je původně adaptivní imunitní systém nalezen u prokaryotních organismů, který, je schopen cíleného odstranění cizích invazivních genetických elementů, jakými jsou plazmidy nebo bakteriofágy. Od té doby, kdy byla objevena možnost jednoduše tenhle systém programovat, Cas proteiny (asociovány s CRISPR) a obzvláště Cas9 byly úspěšně použity v celé řadě aplikací na poli jak základního, tak aplikovaného výzkumu. Kvůli preciznosti, s jakou je tento systém schopen štěpit cílovou sekvenci, byly metody založené na CRISPR/Cas v poslední době využity taky na studium komplexních a repetitivních oblastí v genomech několika druhů. V této diplomové práci jsme zkoumali možnost využití Cas9 proteinu ze Streptococcus pyogenes na izolaci a následnou charakterizaci dvou takových oblastí nacházejících se v nerekombinující části chromosomu Y u dvoudomé rostliny Silene latifolia.

Abstract:

CRISPR/Cas is originally an adaptive immune system found in prokaryotes, that is capable of targeted removal of invading foreign genetic elements, such as plasmids or bacteriophages. Since the discovery of its easy programmability, Cas (CRISPR associated) proteins and especially Cas9 have been successfully used in various applications in the field of basic research and applied science as well. Due to its precise DNA targeting ability, CRISPR/Cas9-based methods have been lately also used to study complex and repetitive regions in genomes of several species. In this diploma thesis, we examine the possibility of using Cas9 protein from Streptococcus pyogenes to isolate and subsequently characterize two such regions located on non-recombining part of Y chromosome in dioecious plant Silene latifolia.
