Study of biologically active substances in grapevine seeds and their influence on selected biochemical markers in Rattus norvegicus

Abstract:

Předložená disertační práce se zabývá studiem antioxidačních vlastností u extraktů ze semen révy vinné (Vitis vinifera, L.) pomocí spektrometrických a chromatografických technik a studiem vlivu těchto semen na Rattus norvegicus, kteří byli vystaveni působení těžkého kovu nebo měli indukovanou obezitu. Pro studium bylo vybráno deset odrůd révy vinné (Cerason, Laurot, Kofranka, Tramín červený, Hibernal, Frankovka, Zweigeltrebe, Erilon, Pálava a Ryzlink vlašský), jejichž semena byla získána z ročníků 2015, 2016 a 2017. Spektrofotometricky, pomocí čtyř principielně rozdílných metod (DPPH●, ABTS●+, FRAP, FR), byla stanovena antioxidační aktivita a pomocí Folin-Ciocalteovi metody byl stanoven obsah celkových polyfenolických sloučenin. V roce 2015 byl u semen z těchto odrůd pomocí HPLC/UV-VIS stanoven obsah 14 antioxidantů a 10 antioxidantů pomocí HPLC s MS detekcí. U odrůdy Cerason byl obsah antioxidantů pomocí HPLC/UV-VIS sledován v průběhu všech tří let 2015 – 2017 a z této odrůdy byl vyroben lyofilizovaný extrakt. Dále byl sledován vliv tohoto extraktu na pokusná zvířata Rattus norvegicus. V čtyř-týdenním experimentu byly práce zaměřeny na sledování protektivního účinku extraktu vůči poškození organismu kadmiem. Byla sledována akumulace kadmia a biochemické markery (antioxidační status, kreatinin, močovina, ALT, AST, superoxiddismutáza, kataláza, metallothionein), v krvi, játrech a ledvině. V šesti-týdenním experimentu byl sledován vliv přídavku extraktu na markery experimentálních zvířat, která měla obezitu. Ve vzorcích krve byl analyzován obsah glukózy, celkového cholesterolu, triacylglyceridů, HDL a LDL. Výsledky této práce poukázaly na vysoký obsah antioxidačních komponent v hroznových semenech a na rozdíly v obsahu u jednotlivých odrůd a v jednotlivých letech. Bylo zjištěno, že semena mají protektivní vliv vůči příjmu těžkých kovů do organismu a pozitivní vliv na lipidový metabolismus.

Abstract:

This thesis deals with antioxidant properties of grape seed extracts (Vitis vinifera, L.) by means of spectrometric and chromatographic techniques and their influence on Rattus norvegicus exposed to heavy metal or induced obesity. Ten grape varieties (Cerason, Laurot, Kofranka, Red Traminer, Hibernal, Frankovka, Zweigeltrebe, Erilon, Pálava and Riesling) were selected for study, whose seeds were obtained from the years 2015, 2016 and 2017. Antioxidant activity was determined spectrophotometrically using four principally different methods (DPPH●, ABTS●+, FRAP, FR) and the total polyphenolic compounds were determined by Folin-Ciocalteu method. In 2015, 14 antioxidants and 10 antioxidants were determinated by HPLC with MS detection. For the Cerason variety, the content of antioxidants was monitored by HPLC / UV-VIS during all three years 2015-2017, and a lyophilized extract was produced from this variety. Furthermore, the effect of this extract on Rattus norvegicus animals was studied. In a four-week experiment, the work focused on monitoring the protective effect of the extract against cadmium damage. Accumulation of cadmium and biochemical markers (antioxidant status, creatinine, urea, ALT, AST, superoxide dismutase, catalase, metallothionein), in blood, liver and kidney were monitored. In a six-week experiment, the effect of extract addition on the markers of experimental animals that had induced obesity was studied. Blood samples were analyzed for glucose, total cholesterol, triglycerides, HDL and LDL cholesterol. The results of this work pointed to the high content of antioxidant components in grape seeds and to differences in content in individual varieties and in individual years. It has been found that seeds have a protective effect on the intake of heavy metals into the body and a positive effect on lipid metabolism.
