Phytoalexine determination in grape vine cell suspension

Abstract:

Fytoalexiny jsou látky syntetizované rostlinou jako součást obranné reakce rostlin vůči abiotickému nebo biotickému stresu. Tuto reakci jsou schopné vyvolat pouze látky, které se nazývají elicitory. Elicitory jsou často součástí těl patogenních mikroorganismů (například ergosterol), nebo fragmenty buněčných stěn samotných rostlin (například oligogalakturonidy). Nejdůležitějšími fytoalexiny u vinné révy jsou stilbeny (resveratrol a jeho deriváty). Použití elicitorů k imunizaci rostlin vůči patogenům by mohla být šetrnější a ekologičtější cesta než doposud používané pesticidy. Docházelo by ke zvyšování koncentrací obranných metabolitů přirozenou a tedy zdravotně nezávadnou cestou. Cílem práce bylo detekovat tyto fytoalexiny v buněčných suspenzích vinné révy (Vitis vinifera cv. Gamay) po působení elicitorů ergosterolu, oligogalakturonidů a jejich kombinace. Stanovení probíhalo pomocí vysokoúčinné kapalinové chromatografie s reverzní fází a určení jednotlivých stilbenů bylo provedeno na základě fluorescence, znalosti retenčních časů a charakteristických absorpčních spekter. Práce byla prováděna ve třech nezávislých opakováních vždy s čtyřdenní buněčnou suspenzí. Elicitace byla prováděna zásobními roztoky elicitorů ergosterolu do výsledné koncentrace 1µM a oligogalakturonidů do koncentrace 0,5 mg/ml. Během práce byly detekovány fytoalexiny resveratrol, viniferin a piceid. Výsledky potvrdily hypotézu synergistického působení ergosterolu a oligogalakturonidů na syntézu stilbenů resveratrolu a viniferinu v buněčných suspenzích vinné révy. Hladina piceidu se po celou dobu experimentu nijak výrazně neměnila, což napovídá, že se může jednat o konstitutivní zásobní formu.

Abstract:

Phytoalexins are substances which are synthetized as a part of plant defense response to abiotic or biotic stress. This response is only induced by substances called elicitors. These elicitors are often part of the pathogen body (for example ergosterol) or fragments of plant cell walls (for example oligogalacturonides). The most important phytoalexins of grapevine are stilbenes (resveratrol and its derivates). The use of elicitors for immunization of plants against pathogens could be more ecological and environmentally friendly way compared to commonly used pesticides. This alternative would allow concentrations of defense metabolites to increase by natural and thus more health-safe way. The aim of this research work was to detect phytoalexins in cell suspensions of grapevine (Vitis vinifera cv. Gamay) previously treated with elicitors: ergosterol, oligogalacturonides and their combination. Determination was carried out using HPLC with reverse phase, fluorescence and the knowledge of retention time, and characteristic absorption spectra. The work was carried out in three independent experiments, each with four-day cell suspensions. Elicitation was performed by stock solution of ergosterol to a final concentration of 1μM and oligogalacturonides to final concentration of 0,5 mg/ml. Elicitation by their combination was carried out by a combination of equal amount of both elicitors. During the experiment, the levels of phytoalexins resveratrol, viniferin and piceid were monitored. The results confirmed the hypothesis of synergistic effect of ergosterol and oligogalacturonides to synthesis of stilbenes, resveratrol and viniferin, in grapevine cell suspension. The level of piceid did not change significantly thought during the experiment, suggesting that piceid could be constitutive stilbene form.
