Nuclear topography and transcriptional activity of the c-myc gene during enterocytic differentiation

Abstract:

Modelovou linii buněk střevního adenokarcinomu HT-29, schopnou diferencovat do enterocytů, jsme použili k analýze jaderného uspořádání c-myc genu a c-myc transkripčních míst. Dále jsme zkoumali prostorové interakce těchto struktur s jadernými doménami, které jsou zahrnuté v transkripčních procesech. Využitím hybridizačních technik s fluorescenčně značenými sekvencemi RNA a DNA v kombinaci s imunocytochemickou analýzou jsme zjistili, že změny v transkripční aktivitě, které souvisí s diferenciačním procesem, jsou doprovázeny přeuspořádáním chromatinu v interfázním jádře. To se projevuje specifickou jadernou a teritoriální redistribucí transkripčně aktivních a neaktivních c-myc genů. Transkripce c-myc genů se vyskytovala ve střední části interfázního jádra a obvykle asociovala s okrajem centrálně umístěného jadérka. Diferenciační procesy neměly vliv na polohu c-myc transkripčních míst. Kromě toho jsme zjistili, že se u většiny buněk transkripční místa c-myc genů překrývají s místy, kde se vyskytuje RNA polymeráza II, a že se nacházejí v těsné blízkosti těch jaderných skvrn "nuclear speckles", které jsou umístěné ve vnitřní části buněčného jádra. Na základě zmíněných zjištění usuzujeme, že existuje blízký vztah mezi strukturou chromatinu a jeho funkcí. Přičemž funkce chromatinu je regulována faktory, které jsou obsaženy v jaderných doménách, které s chromatinem úzce asociují.

Abstract:

The nuclear arrangement of the c-myc gene and the c-myc transcription site, as well as the spatial interaction of these structures with the nuclear domains involved in RNA synthesis and processing, was analyzed using the enterocytic differentiation model of HT 29 cells. Our experiments, using sequential RNA/DNA fluorescence in situ hybridization combined with immunocytochemistry, revealed that differentiation-related changes in transcription activity are accompanied by the reorganization of chromatin within the interphase nuclei. This manifests itself as a specific nuclear and territorial redistribution of transcriptionally active and inactive c-myc genes. C-myc transcription usually took place in a central part of interphase nucleus and was preferentially associated with the periphery of the centrally located nucleoli. The differentiation process did not influence the positioning of c-myc transcription sites. Furthermore, we have found that a single transcription site of the c-myc gene co-localized with RNAP II positive domains and in a majority of cells, the c-myc transcription site was observed in close proximity to one of the nuclear speckles, located in the internal parts of the cell nucleus. On the basis of these findings, we suggest the existence of a close inter-relationship between chromatin structure and function, which is additionally regulated by factors involved in chromatin associated domains.
