The use of transparent electrodes for construction of biosensors

Abstract:

Tato diplomová práce byla zaměřena na využití transparentních elektrod pro konstrukci biosensorů. V první části byly využity již použité čipy (pokryté 50 nm silnou vrstvou zlata), které jsou primárně určené pro metodu rezonance povrchového plasmonu (SPR), k elektrochemické detekci vazby HSA a protilátky anti-HSA AL-01. Kromě této interakce byla také detekována vazba modelového organismu Escherichia coli na předem imobilizovaný zlatý povrch se specifickou protilátkou. Obě tyto interakce byly zaznamenávány pomocí amperometrie a připravených imunokonjugátů obsahujících enzym HRP. V druhé části této práce byly pomocí SPR prozkoumány jednotlivé možnosti úpravy povrchů měřících čipů. Bylo využíváno několika druhů čipů potažené rozdílným povrchem. Měření bylo prováděno pomocí dvou systémů od různých výrobců (zařízení Plasmon VI z Ústavu fotoniky a elektroniky AV ČR a zařízení SPRNavi™ 210A od finské firmy Bionavis) a došlo ke srovnání jednotlivých způsobů imobilizace a celkové odezvy. Jako modelový organismus byla i v těchto měřeních použita bakterie Escherichia coli.

Abstract:

This thesis was focused on transparent electrodes and their using for construction of biosensors. In the first part of this thesis, used chips (covered by 50 nm layer of gold), which are primary apply for surface plasmon resonance, were using for electrochemical detection of interaction between HSA and antibody anti-HSA AL-01. Besides this measurement, the interaction of model organism Escherichia coli with a gold surface modified with a specific antibody were investigated. Both interactions were detected by amperometry and there were used imunoconjugates with enzyme HRP. In the second part of this thesis, possibilities of treatment the surfaces of chips were investigated by method of SPR. There were used several kind of chips covered by a different surfaces. Measurements were done with two systems from different producer (device Plasmon VI from Institut of Photonics and Electronics AS CR and device SPRNavi™ 210A by Finnish company Bionavis) and comparing of different types of immobilization and overall response were created. Escherichia coli was also used in these measurements like a model organism.
