Monitoring Of Changes In DNA Methylation As Prognostic Biomarkers In Cervical Cancer

Abstract:

Cílem této diplomové práce je nalezení potenciálních diagnostických a prognostických markerů pro karcinom děložního čípku založeny na analýze metylace DNA s možností vyšetřovat je za použití neinvazivních metod. Teoretická část práce představuje problematiku karcinomu děložního čípku. Následuje shrnutí jednotlivých epigenetických mechanismů s podrobnějším popisem metylace DNA a jejího významu v karcinogenezi. Na konci této části je zmíněn přehled molekulárních a epigenetických biomarkerů pro karcinom děložního čípku. V experimentální části, která probíhala v Ústavu klinické biochemie a diagnostiky ve Fakultní nemocnici v Hradci Králové, jsme si nejprve navrhli primery pro geny zvolené na základě literární rešerše. Optimalizovali jsme metody kvantitativní PCR (qPCR) a metylačně-specifickou analýzu křivek tání s vysokým rozlišením (MS-HRM), které jsme použili pro posouzení rozsahu metylace vybraných genů u našich vzorků. Jednalo se o vzorky odebrané z děložního čípku s nádorovým bujením a nenádorové kontrolní vzorky bez diagnózy nádorového bujení. V závěru práce je na základě statistické analýzy dat uvedena korelace vybraných genů se zaznamenanými klinicko-patologickými charakteristikami karcinomu děložního čípku.

Abstract:

The diploma thesis is devoted to finding potential diagnostic and prognostic markers for cervical cancer, which would be based on DNA methylation analysis and could be investigated using non-invasive methods. The experimental part of the diploma thesis took place at the Institute of Clinical Biochemistry and Diagnostics in Hradec Králové.The theoretical part of the work presents the topic of cervical cancer. The following is an introduction to individual epigenetic mechanisms with a more detailed description of DNA methylation and its importance in carcinogenesis. An overview of molecular and epigenetic biomarkers for cervical cancer is mentioned at the end of this section. In the experimental part, we designed primers for genes selected based on a literature search. We optimized quantitative PCR (qPCR) methods and high-resolution methylation-specific melting curve (MS-HRM) analysis, which we used to determine the extent of methylation of selected genes in our samples. These samples were taken from the cervix with tumor growth and non-tumor control samples without a malignant diagnosis. At the end of the work, the correlation of selected genes with the recorded clinicopathological characteristics of cervical cancer is presented.
