Development and Validation of ELISA Kit for Determination of Allergic Proteins from Egg White

Abstract:

Dizertační práce se zabývá vývojem a validací imunoanalytické soupravy pro detekci a kvantifikaci alergenních proteinů vaječného bílku v potravinách metodou ELISA (Enzyme-Linked Immunosorbent Assay). Úvodní část v teorii je věnována potravinovým alergiím a popisu jednotlivých potravinových alergenů. Další část popisuje analytické metody, které potravinové alergeny detekují a stanovují jejich množství v testované potravině. Tyto metody jsou rozděleny na fyzikálně-chemické, imunochemické a molekulárně-biologické. Třetí část se zaobírá validací imunoanalytických metod, legislativním základem validace a experimentálním a statistickým postupům ve validačním procesu. Závěrečná část popisuje legislativní vymezení problematiky potravinových alergenů. Experimentální část zahrnuje použité chemikálie, pomůcky a přístroje. Podrobně popisuje postup imunizace zvířat a vývoj polyklonální protilátky proti alergenním proteinům vaječného bílku, její izolaci a testování. Dále se zaměřuje na vývoj a optimalizaci immunoassay. V závěru se zabývá validací nově vyvinuté soupravy. Ve výsledcích a diskuzi jsou shrnuty veškeré získané výsledky a poznatky. První část je věnována sledování imunitní odpovědi imunizovaných zvířat, izolaci protilátky a jejímu testování. V druhé části jsou zaznamenány výsledky vývoje a optimalizace metody ELISA pro kvantitativní stanovení alergenních proteinů vaječného bílku. Třetí závěrečná část popisuje validaci vyvinuté soupravy, jež je zakončena mezilaboratorním porovnávacím testem s účastí jedenácti odborných pracovišť.

Abstract:

ELISA method (Enzyme-Linked Immunosorbent Assay). The initial part of the Theoretical section is devoted to food allergies and describes various food allergens. The part that follows describes analytical methods for detection and quantification of food allergens in tested food sample. These methods are divided to physical ? chemical methods, immunochemical methods and molecular ? biological methods. The third part is concerned to validation of immunoanalytical methods, the legislative basis of the validation and experimental and statistical procedures in validation process. The concluding part of the Theoretical section discusses the legislative definition of food allergens problem. The Experimental section includes used chemicals, tools and instruments. This part describes in detail the procedure of immunisation of animals and development of polyclonal antibody against allergic proteins of egg white and their isolation and testing. The part that follows is focused on the development and optimization of immunoassay. The concluding part of the Experimental section describes the validation newly developed kit. The section Results and Discussion summarises all the results and findings obtained. Its first part deals with the immune response of immunised animals, isolation of antibody and their testing. The second part concerns results of the development and the optimization of ELISA method for the quantitative determination of egg white proteins. The third and final part is focused on the validation of the newly developed kit which is finished by interlaboratory study with the participation of eleven laboratories.
