The effect of histamine on the expression of anti-apoptotic and pro-apoptotic proteins in human neutrophils activated with different stimuli

Abstract:

Neutrofilní granulocyty jsou prvními buňkami, které se objevují v místě zánětu a jejich hlavní funkcí je zneškodnění cizorodého patogenu. Využívají přitom proteolytické enzymy a reaktivní metabolity kyslíku. Aktivované neutrofily hynou v procesu programované buněčné smrti – apoptózy. Následně jsou neutrofily fagocytované makrofágy a dochází k rezoluci zánětu. Pokud se v organismu vyskytne zánět, dojde k produkci zánětlivých mediátorů, které jsou uvolněny z granulí žírných buněk a bazofilů. Primárním mediátorem zánětu je histamin a jeho hlavní funkcí je regulovat tento proces. Cílem bakalářské práce bylo objasnit vliv histaminu na expresi antiapoptického proteinu Mcl-1 a proapoptického proteinu Bax v lidských neutrofilech aktivovaných stimuly oxidativního vzplanutí OZP (opsonizovaný zymosan) a PMA (Forbol-12-myristát-13-acetát). Z provedených experimentů jsme vliv histaminu neprokázali. Ze zjištěných výsledků pouze vyplývá, že použité stimuly zvyšují expresi proapoptického proteinu Bax ve zvolených časových intervalech.

Abstract:

Neutrophilic granulocytes are the first cells at the site of inflammation, and their primary function is destruction of heterogeneous pathogens by using proteolytic enzymes and reactive oxygen metabolites. Activated neutrophils are cleared in the process of programmed cell death - apoptosis. Subsequently they are phagocytised by macrophages which leads to resolution of inflammation. If there is inflammation in the body, the mediators of inflammation are produced. These are released from the granules of mast cells and basophils. The primary mediator of inflammation is histamine and its main function is to regulate this process. The aim of the thesis was to clarify the effects of histamine on the expression of anti-apoptotic protein Mcl-1 and pro-apoptotic protein Bax in human neutrophils activated with stimuli of the oxidative burst OZP (Opsonized Zymosan Particles) and PMA (phorbol-12-myristate-13-acetate). In our experiments we did not prove the influence of histamine. On the other hand, we found out that the stimuli we used increased the expression of pro-apoptotic protein Bax in the chosen time intervals.
