Evaluation of biofilm formation by arcobacters

Abstract:

Teoretická část diplomové práce se zabývá tvorbou bakteriálního biofilmu, způsoby jeho odstranění a především pak metodami detekce. Dále jsou obsaženy kapitoly, které se zabývají charakteristikou rodů Arcobacter spp. a Campylobacter spp., způsoby přenosu a klinickými projevy onemocnění. V části experimentální jsou shrnuty použité metody detekce biofilmu a metody pro rozlišení živých a mrtvých buněk. Pomocí vybraných metod byla sledována tvorba biofilmu u několika kmenů rodu Arcobacter. Biofilm byl detekován pomocí Christensenovy metody v mikrotitrační destičce za různých podmínek ve vybraných médiích. Stanovení kultivací na kuponech z nerezové oceli bylo provedeno stěrovou metodou. Dále byla zavedena a zoptimalizována metoda EMA/PMA-PCR pro odlišení živých a mrtvých buněk.

Abstract:

The theoretic part of thesis concerns with the formation of bacterical biofilm, methods for its remove and primarily with methods for its detection. There are the chapters about basic characteristics of the genus Arcobacter spp. and Campylobacter spp., about abilities of its transfer to the human and about the clinical aspects of diseases. In the experimental part are summarized methods for detection of biofilm, methods for distinguishing of died and viable cells. The formation of biofilm was monitored using methods for several species of the genus Arcobacter. Biofilm was determined using the Christensen´s method in the polyethylene microtitrate plate. This determination was done in the selected dilutions and by various conditions. The culture assay on the stainless steel coupons was done by swab method. The EMA/PMA-PCR that distinguish died and viable cells was optimized.
