Elastic Liposomes for Encapsulation of Canabinoids

Abstract:

Elastické liposomy jsou díky své deformovatelné struktuře schopny enkapsulovat velké množství aktivních látek a usnadňovat jejich transport kůží až na cílová místa. Kanabionoidy jsou významnými látkami při léčbě řady onemocnění. Jejich omezením je však špatná biologická dostupnost a náchylnost k degradaci. Pro překonání těchto nedostatků je vhodnou strategií jejich enkapsulace do systému elastických lipozomů. V rámci této diplomové práce byly připraveny formulace elastických lipozomů ze sojového fosfatidylcholinu metodou hydratace tenkého filmu. Jako aktivátory hran byly použity Tween80 a D-alfa-Tocopherol polyethylene glycol 1000 sukcinát. Enkapsulovanou terapeuticky aktivní látku představoval kanabidiol. U připravených formulací byly charakterizovány fyzikálně chemické vlastnosti včetně velikosti vezikul, distribuce a zeta potenciálu. Rovněž byly zhodnoceny z hlediska stability, enkapsulační účinnosti, antioxidační aktivity, cytotoxicity a na závěr schopnosti transdermální penetrace ex vivo.

Abstract:

Due to their deformable structure, elastic liposomes can encapsulate large amounts of active substances and facilitate their transport through the skin to the target sites. Cannabinoids play important role in the treatment of variety diseases. However, their limitation is their poor bioavailability and susceptibility to degradation. To overcome these shortcomings, their encapsulation in the elastic liposomes is an effective strategy. In the thesis, the thin film hydration method was used to prepare elastic liposome formulations containing soy phosphatidylcholine. Tween80 and D-alpha-Tocopherol polyethylene glycol 1000 succinate were used as edge activators. The encapsulated therapeutically active substance was cannabidiol. The physicochemical properties of the prepared formulations were examined, including vesicle size, distribution and zeta potential. The samples were also evaluated for stability, encapsulation efficiency, antioxidant activity, cytotoxicity and finally ex vivo transdermal penetration ability.
