MS analysis of proteins with posttranslational modifications with a focus on phosphorylation

Abstract:

Posttranslační modifikace, které jsou přítomné na proteinech a peptidech, jsou nositeli mnoha funkcí, například se může jednat o signalizaci uvnitř buňky nebo vliv na strukturu a tvar samotného proteinu, který úzce souvisí s jeho funkcí a rovněž s odbouratelností v buňce, nebo mimo ni. Posttranslačními modifikacemi se dnes zabývá, mezi jinými, vědní obor proteomika. Proteomická analýza je různorodá směsice přístupů a strategií k přípravě vzorku a jeho analýze. Tato práce se konkrétně zabývá studiem fosforylací a využívá přístup přípravy vzorku afinitní chromatografií na oxidu titaničitém (MOAC), který je zde používán ve formě několika nosičů. Analýza z takto získaných vzorků probíhala na hmotnostním spektrometru LTQ-Orbitrap XL s MALDI ionizací a Q Exactive s nano-LC-ESI. Součástí této práce je testování vlastností nosičů (Titansphere, magnetické "core-shell" nanočástice a magnetické nanotrubice z oxidu titaničitého, které jsou potaženy magnetickými Fe3O4 nanočásticemi). Oba mag. nosiče byly vytvořeny v laboratořích FCHT-Univerzity Pardubice.

Abstract:

Post-translational modifications of proteins and peptides have many functions, for example it may be cell signaling, or an effect on the structure and shape of the protein itself that is closely related to its function as well as its degradation within or outside the cell. In nowadays post-translational modifications are the object of proteomical studiing, amongst others. Proteomic analysis is a diverse combination of approaches and strategies for sample preparation and analysis. This diploma thesis deals specifically with the study of phosphorylation and uses preparation of the sample by affinity chromatography on titanium oxide (MOAC), which is used in the form of several carriers. Analyse of these samples was carried out on an LTQ-Orbitrap mass spectrometer analyzer with MALDI ionization and Q Exactive mass spectrometer with nano-LC-ESI. Part of this work is an evaluation of the properties of carriers (Titansphere, magnetic core-shell nanoparticles and magnetic nanotubes of titanium dioxide covered with magnetic Fe3O4 nanoparticles). These magnetic carriers were prepared in the FCHT laboratories of the University of Pardubice.
