The Use of Fluorescence Spectrophotometry Methods in Colloidal Chemistry

Abstract:

Fluorescence je jev, kdy látka vysílá po ozáření do prostoru světlo. Záření, které fluorescenci vyvolává, se nazývá excitační, záření vysílané látkou se nazývá emisní. Fyzikální podstata fluorescence spočívá ve vlastnostech elektronového obalu atomů v molekulách fluorochromu. Fluorescenční metoda, která se používá ke stanovení vzorků na základě spektra pohlceného světla, se nazývá fluorescenční spektrofotometrie. Fluorescenční spektrofotometrie se využívá k výzkumu materiálů v biofyzice, biochemii, biomedicíně a dalších odvětvích. Tato práce je zaměřena na měření excitačního a emisního spektra zásobních roztoků fluoresceinů (fluorescein sodium, fluoresceinamin a fluorescein isothiocyanate-dextran), při různých koncentracích a vlnových délkách a na sledování konformačních změn kyseliny hyaluronové a karboxymethylcelulózy pomocí fluorescenčního spektrofotometru Schimadzu RF-1501. Přístroj měří fluorescenční spektra v rozsahu 220 nm až 900 nm a umožňuje automatické vyhledání optimální vlnové délky pro excitaci a emisi záření. Měření vzorků na fluorescenčním spektrofotometru sleduje závislost intenzity fluorescence na koncentraci rozpuštěné látky. Jako rozpouštědlo byla použita deionizovaná voda, ethanol pro měření na UV-VIS spektrofotometru a aceton pro měření na fluorescenčním spektrofotometru. Dále bylo provedeno měření s přidáním kyseliny hyaluronové a karboxymethylcelulózy a s úpravou pH roztoků prostřednictvím NaOH a HCl. Byla měřena závislost intenzity fluorescence na vlnové délce. Z naměřených hodnot byly sestaveny grafy závislosti intenzity exitačních a emisních spekter na pH prostředí. Tato měření umožnila sledování konformačních změn biopolymerů v závislosti na pH. Bylo zjištěno, že koncentrace zásobních roztoků fluoresceinů není přímo úměrná intenzitě naměřených píků, ale na základě naměřeného spektra a získání bodu zlomu byla potvrzena konformační změna statistické klubko/šroubovice hyaluronanu a karboxymethylcelulózy se všemi použitými druhy fluoresceinů.

Abstract:

Fluorescence is a phenomenon where a substance emits light, after radiation, into space. The radiation that fluorescence evokes is called "excitation" and the radiation emitted by asubstance is called "emission". The physical principle of fluorescence is based upon the characteristics of the electron shells of atoms in fluorochromemolecules. The fluorescence method used to determine samples on the basis of the spectra of absorbed light is called Fluorescence Spectrophotometry; and is used for materials research in Biophysics, Biochemistry, Biomedicine and other fields. This work is oriented on measuring the excitation and emission spectraof stock fluorescein solutions (e.g. fluorescein natrium, fluorescein amine and fluorescein isothiocyanate-dextran) in different concentrations and wavelengths to monitor the conformational change of the hyaluronic acid and carboxymethylcelluloze with the help of a Schimadzu RF-1501 Fluorescence Spectrophotometer. This instrument measures fluorescence spectrums in the 220 nm to 900 nm range and can automatically find the optimal wavelength for the excitation and emission of radiation. Sample measurement on a fluorescence spectrophotometer monitored the dependence of fluorescence intensity on the concentration of a dissolved substance. Deionized water, ethanol for measurement on a UV-VIS Spectrophotometer and acetone were used as solventsfor fluorescence spectrophotometer measurements. Furthermore, measurements were performed with the addition of hyaluronic acid and carboxymethylcellulose and adjusted by pH solutions using NaOH and HCl. The dependence fluorescence intensity on wavelength was also measured. Dependence charts intensity excitation and emission spectra on the pH were compiled from the measured values. These measurements allowed monitoring conformational changes of biopolymers depending on the pH. It was found that the concentration of stock fluorescein solutions is not directly proportional to the intensity of plotted peaks, but rather, on the basis of the measured spectrum and discovering the breakpoint, the conformational change statistical clue/helix of hyaluronic acid and carboxymethylcellulose with all kinds of fluorescein was confirmed.
