Sequence analysis to predict the possibility of the use of picornavirus inhibitors against Parechoviruses, Kobuviruses, Cardioviruses, and Cosaviruses

Abstract:

Pikornaviry jsou malé, ikosaedrální viry způsobující mnohé závažné onemocnění lidí i zvířat. Zatím není k dispozici žádný lék pro léčbu pikornavirových infekcí i přesto, že u některých látek byla prokázána antipikornavirová aktivita. Krystalové struktury řady pikornavirů byly vyřešeny a to usnadňuje návrh léčiv proti těmto virům. Avšak struktury mnohých pikornavirů včetně lidských parechovirů, kobuvirů, kardiovirů a kosavirů zatím vysvětleny nebyly. Nicméně, sekvenční analýzy nám mohou umožnit odhadnout, zda je možné proti těmto virům použít inhibitory studované v souvislosti s pikornaviry se známou strukturou. Cílem této práce je podat přehled o pikornavirech, jejich životním cyklu a látkách, u kterých byla prokázána antipikornavirová aktivita a podle sekvenčních analýz odhadnout možnost jejich použití proti pikornavirům se zatím neznámou strukturou.

Abstract:

Picornaviruses are small, icosahedral viruses which cause many severe diseases of humans and animals. To date there is no treatment for picornaviral infections available although some compounds have been shown to have antipicornaviral activity. Crystal structures of series of picornaviruses have been solved and it facilitates the drug design against these viruses. But there are still many viruses including human parechoviruses, kobuviruses, cardioviruses and cosaviruses, whose structures need to be elucidated. However, sequence analyses of viral proteins might allow us to predict the possibility of the use of inhibitory compounds in respect to picornaviruses with known structure against viruses with unknown structure. The aim of this thesis is to give an overview about picornaviruses, their life cycle and compounds which have shown antipicornaviral activity and to predict their use against picornaviruses with unknown structure according to sequence analyses.
