Abstract:

Chronická onemocnění některých urgánů ( např. játra, slinivka břišní ) jsou charakterizována ztrátou vlastního funkčního parenchymu, který je souběžně nahrazován ukládaným novotvořeným vazivem. Fibroprodukce je často velmi progresivní a vede k morfologické a funkční alteraci daného orgánu. Za velkou většinu fibroprodukce jsou v játrech a slinivce břišní zodpovědné buňky s charakteristikami myofibroblastů. S ohledem na jejich buněčný tvar jsou nazvány hvězdicovité buňky ( stellate cells, SC). Vyskytují se ve dvou základních fenotypových formách- klidové a aktivované. Pro možnost regulace patologicky navýšené fibroprodukce při chronických orgánových onemocněních, je třeba především regulovat činnost aktivované formy hvězdicovitých buněk. Experimenty popisované v disertační práci objektivizují přítomnost hvězdicovitých buněk pankreatu ve vzorcích resekátů pankreatu člověka.Na kultuře hvězdicovitých buněk pankreatu laboratorního potkana je ověřena jejich responzibilita vůči cytokinům se známým biologickým účinkem na tyto buňky ( např.navýšená proteosynteza- fibroprodukce). V hlavním experimentu PhD práce je ověřena přítomnost IGF II receptoru na povrchu hvězdicovitých buněk a navýšení jeho exprese, jako stacionární součást aktivace hvězdicovitých buněk. Tato vlastnost je vhodná pro využití při buněčné adresaci antifibrotické terapie cíleně a vysoce selektivně právě pro aktivovanou formu ( vysoce proteosynteticky aktivní) hvězdicovitých buněk pankreatu.

Abstract:

Chronical organ diseases ( of f.e. liver, pancreas) are accompanied by the loss of functional parenchyma, which is followed by and replaced by newly produced collagen fibres tissue. Fibroprodution is ussually very progressive and leads towards the morphological and functional changes of this organ. In liver and pancreas , the specific subunit of myofibroblast-iike cells are responsible for the major part of extracellular matrix oveproduction there. According to their cellular shape they are named stellate cells. They are present in two fenotypic forms - quiescent and activated. For the purpose to regulate the patological collagen fibres tissue overproduction in such organ diseases, there is necessity to regulate the activated form of stellate cells. The experiments presented in the PhD thesis demonstrate the presence of human pancreatic stellate cells in the samples of human pancreatic parenchyma. In the culture of rat pancreatic stellate cells there are demostrated the specific stellate cells reactions to the cytokines with known biological effects, on their activity ( increased proteosyntetic activity - fibroprodution). In the main PhD thesis experiment there is proved the presence of IGF II receptor on the surface of rat pancreatic stellate cells in culture; and the increased expression of IGF II receptor, as the constant feature following the actiovation of these cells. This characteristic is suitable for the cellular targeting technique, when we would like to deliver – highly selectively - some antifibrotic substance to these activated pancreatic stellate cells.
