Physiology and pathology of infection in bats

Abstract:

Netopýři jsou přirozeným hostitelem mnoha patogenů díky jejich specifické fyziologii, která jim umožňuje přežít infekci bez související patologie. Během svého životního cyklu mění netopýři výrazně tělesnou teplotu od aktivního letu k hibernaci. Tyto změny teploty představují další selekční tlak způsobující změny v jejich fyziologii. Známky adaptivní evoluce vzniklé jako reakce na fyziologické změny a infekci jsou detekovatelné v netopýřích genomech. Adaptace na tyto selekční tlaky je možné studovat pomocí transkriptomických a genomických metod. Cílem této práce je popsat adaptace k endemickým netopýřím nákazám přítomné v genomech netopýrů. Studovala jsem fyziologické adaptace vzniklé evoluční odpovědí na hibernaci a specifika imunitní reakce netopýrů během hibernace. Výsledky prezentované v této práci poukazují na adaptace netopýrů vzniklé koevolucí s ancestrálně Palearktickou plísní Pseudogymnoascus destructans. Pomocí metody založené na kandidátních genech byly detekovány geny ovlivňující imunitní systém, hojení ran a integritu kůže pod pozitivní selekcí. Adaptace vzniklé na základě infekcí jsou u netopýrů podpořeny adaptacemi, které vznikly jako evoluční odpověď na změny tělesné teploty během hibernace. Tyto adaptace chránící netopýry během hibernace mohou mít pozitivní vliv i na reakci netopýrů na přítomnost infekce. Netopýři druhu Myotis myotis vykazují adaptace na různé hibernační podmínky v genech pro proteiny s funkcí molekulárních chaperonů. Tyto adaptace získané evoluční odpovědí na hibernaci mohou společně s adaptacemi imunitního systému ovlivnit i netopýří odpověď na infekce. Transkriptomická studie prezentovaná v této práci poukazuje na potlačenou reakci buněk na infekci lyssaviry při experimentální infekci buněk kultivovaných při nízkých teplotách. Buněčná odpověď v euthermii naopak ukazuje zvýšenou produkci transkriptů souvisejících s TGF-β protizánětlivou signalizací, která může vést k toleranci. Výsledky práce jsou tak konzistentní s recentním výzkumem v této oblasti. Koexistence netopýrů a jejich endemických patogenů je tak umožněná souhrou mechanismů, které chrání tkáně před poškozením a silnou regulací imunitní reakce během infekce.

Abstract:

Bats host a number of endemic pathogens, but they have evolved physiology that enables them to harbor pathogens without developing the resulting pathology. Bats also experience temperature extremes during active flight and hibernation which pose another selective pressure to their physiology. Adaptive evolution to withstand interaction of selective pressures related to rapid physiological changes and infection is imprinted in the bat genomes. Adaptations to the selective pressure of pathogen infection can be studied using genomic and transcriptomic approaches. This thesis aimed to describe the adaptation to endemic infections present in genomes of hibernating bats. I investigated the physiological adaptations to specific hibernation conditions and the bat reaction to viral infection during hibernation. The results presented in this thesis show that the bats in the Palearctic regions, ancestral region of pathogenic fungus Pseudogymnoascus destructans, have coevolved with the fungus. The candidate gene approach used to detect molecular adaptation acquired during the bat coevolution with P. destructans showed positive selection in genes related to immune downregulation, improved wound healing, and the morphological adaptation of skin layer protection. The adaptations to pathogen pressure in hibernating bats are accompanied with adaptive evolutionary changes that bats have acquired in response to the changing body condition during hibernation. The bat physiology pre-adapted to hibernation conditions may also impact the bat reaction to infection. Myotis myotis bats have developed adaptations in genes related to molecular damage repair (i.e. in chaperones) in response to distinct environmental conditions at the hibernation site. The adaptations acquired in response to hibernation can improve bat homeostasis maintenance during infection, supported by multiple adaptations in the immune system. The transcriptomic study presented in this thesis shows lack of reaction to lyssavirus infection during cultivation of bat cells at low temperature. The cellular reaction to the infection in conditions simulating euthermia seems to be heavily affected by TGF-β antiinflammatory signalling, suggesting a response directed towards immune tolerance. These results are consistent with recent research of bat immunity to infectious diseases. Bat coexistence with their endemic pathogens is thus enabled by synergy of tissue protective mechanisms together with strongly regulated immune response to infection.
