Analysis of biologically relevant substances by luminescence sensors

Abstract:

Bioanalýza je v současné době rychle se rozvíjejí oblast chemie, která využívá luminescenční značky pro zvýšení citlivosti daných metod. Předkládaná práce je zaměřena na luminescenční nanokrystalické kvantové tečky. Kvantové tečky byly navázány na biologicky významné látky (proteiny a nukleové kyseliny) a takto syntetizované konjugáty kvantových teček byly charakterizovány pomocí elektromigračních metod. Tato práce představuje sondu, která slouží k detekci specifické sekvence nukleových kyselin a která je založena na principu Försterova resonančního přenosu energie. Dále je studována izotachoforéza jako alternativní způsob přípravy vzorku nukleových kyselin. V práci je také popsána vícekroková syntéza imunosondy značené kvantovými tečkami.

Abstract:

Bioanalysis is recently developing and progressing area, which generally uses luminescent labels in order to increase the sensitivity. This work is focused on quantum dots nanocrystals as luminophores. Quantum dots were conjugated with biologically relevant molecules (proteins and nucleic acids) and conjugates were characterized by electromigration methods. A quantum dot probe is based on Förster resonance energy transfer and it is intended for detection of a specific nucleic acid sequence. Furthermore, isotachophoresis is presented as an alternative way of nucleic acids sample preparation. Finally, an advanced synthetic method of immunoprobe based on quantum dot is described.
