Mechanistic analysis of selected microRNAs on the model of colorectal cancer cell lines

Abstract:

Objectives: Colorectal cancer (CRC) is one of the most common malignancy. Molecular background of CRC is associated with the number of genetic alterations. miRNA are short non-coding RNA that post-transcriptionally regulate gene expression. miR-21 expression levels were up-regulated in the CRC tumors in the most studies performed in this field. This indicate that miR-21 could be one of the key miRNAs in CRC pathogenesis. Methods: In our study we demonstrate the impact of miR-21 on the phenotype of colorectal cancer DLD1 cells through its down-regulation by transient transfection of complementary anti-miR-21 by use of Lipofectamine 2000. DLD1 cell were also treated with common chemotherapeutics (5-fluorouracil, oxaliplatin, irinotecan), and tested on the viability (MTT) and apoptosis: propidium iodide and PARP cleavage. Results: By use of transient transfection of anti-miR-21 we have reached decrease in miR-21 levels to approximately to 25%. This decrease itself has not influenced viability of DLD1 cells, neither their apoptotic rate. Interestingly, we have shown significant sensitizing effect of anti-miR-21 transfection to the treatments of selected chemotherapeutics, mainly 5FU and oxaliplatine. We have observed increase in apoptotic rate up to 30% in anti-miR-21 transfected DLD1 cells in comparison to cells only treated by cytostatics. Conclusion: Our study confirmed significance of miR-21 in colorectal cancer cell line DLD1, mainly on the level of apoptotic effects. Our data, after detailed validation on the different CRC cell lines, could be the rational for the improvement of colorectal cancer therapy by sensitizing of the cells to standard chemotherapeutic regimens.

Abstract:

Východisko: Kolorektálny karcinóm je jedným z najčastejších nádorov spomedzi všetkých nádorových ochorení. Vzniká mnohými genetickými zmenami. miRNA sú krátke nekódujúce úseky RNA, ktoré posttranskripčne regulujú expresiu génov. Hľadina miR-21 bola mnohonásobne zvýšená v nádoroch vo väčšine štúdiách zameraných na CRC. To ju predurčuje ako jednu z kľúčových miRNA v kancerogenéze. Metódy: Štúdia sa snažila preukázať vplyv miR-21 na fenotyp buniek kolorektálneho karcinómu DLD1 pomocou zníženia hladiny transientnou transfekciou komplementárnej anti-miR-21 použitím Lipofectaminu 2000. Bunky boli ošetrené chemoterapeutikami (5-fluorouracil, oxaliplatina a irinotecan) a podrobené testom na viabilitu (MTT) a apoptózu: propídium jodid a štepení PARP. Výsledky: Transientou transfekciou anti-miR-21 sa nám podarilo dosiahnuť poklesu hladiny miR-21 na približne 25 %. Tento pokles sám o sebe nemal vplyv na viabilitu buniek DLD1 ani na podiel buniek podstupujúcich apopózu. Ukázalo sa však, že významne senzitivizuje bunky k účinku vybraných cytostatík, a to predovšetkým k 5FU a oxaliplatine, kde sme u transfekovaných buniek pozorovali až o 33% silnejšiu indukciu apoptózy oproti bunkám ošetrených iba cytostatikom. Záver: Štúdia potvrdila významnosť miR-21 na bunkovej línií kolorektálneho karcinómu, predovšetkým na úrovni apoptotických dejov. Získané poznatky by po dôkladnejších validáciách na ďalších bunkových líniách kolorektálneho karcinómu mohli byť využité pri zvyšovaní účinnosti terapie kolorektálneho karcinómu sensitivizáciou nádoru k štandardným chemoterapeutickým režimom.
