DNA damage evaluation by comet assay method

Abstract:

Metoda comet assay se používá při hodnocení poškození molekul DNA v živých buňkách a díky tomu může být využita i k hodnocení vlivu léčiv a nových lékových forem na poškození DNA především u eukaryotních buněk. Principem metody je migrace a separace řetězců DNA v elektrickém poli v závislosti na jejich délce - tedy míře poškození. Analýza je prováděna na celých buňkách po narušení jejich membrány. Tímto způsobem je v okolí buňky, která tvoří tzv. hlavu komety, tvořen poškozenou DNA tzv. ohon (ocas) komety. Jedním z klíčových parametrů pro úspěšný průběh analýzy je schopnost lyzačního pufru narušit buněčnou membránu. Cílem této bakalářské práce bylo srovnat vhodnost tří typů lyzačního pufru a vliv jeho složení na rozměr komet. Analýza byla provedena na savčích buňkách - myších fibroblastech 3T3. Poškození DNA bylo způsobeno expozicí roztoku peroxidu vodíku. Následně byl zkoumán rozměr ohonu komety. Tedy do jaké míry jsou jednotlivé lyzační pufry schopny narušovat buněčnou membránu v závislosti na jejich složení.

Abstract:

The comet assay method is used in evaluation of damage to DNA molecules in living cells and can therefore also be used to assess the effect of drugs and new dosage forms on DNA damage, especially in eukaryotic cells. The principle of the method is migration and separation of DNA chains in an electric field depending on their length - i.e. the level of damage. The analysis is performed on whole cells after disruption of their membrane. In this way, the tail of the comet is formed by damaged DNA in the surroundings of the cell, which forms the so-called head of the comet. One of the key parameters for a successful analysis is the ability of the lysis buffer to disrupt the cell membrane. The aim of this bachelor thesis was to compare suitability of three types of lysis buffer and influence of its composition on the size of the comets. The analysis was conducted on mammalian cells - mouse fibroblasts 3T3. DNA damage was caused by exposure to hydrogen peroxide solution. Afterwards, the size of the comet tail was investigated. Thus, the extent to which the individual lysis buffers are able to disrupt the cell membrane depending on their composition was explored.
