Structure and function of 14-3-3zeta monomeric mutants

Abstract:

Dimerní charakter proteinů 14-3-3 je často stěžejní pro jejich funkci a pro jejich schopnost vázat fosforylované vazebné partnery. 14-3-3 tvoří homo/heterodimery, které jsou v dynamické rovnováze s monomery. Dimerní forma 14-3-3 proteinů byla dobře prostudována, nicméně struktura a funkce monomerní formy zůstávají nedostatečně popsány. Monomerizace 14-3-3ζ proteinu je regulována různými způsoby, včetně fosforylace Ser58 lokalizovaného na dimerním rozhraní. Fosfomimikující mutant (S58E) proteinu 14-3-3ζ způsobuje pouze částečnou disociaci. Nicméně my jsme navrhli dva mutanty proteinu 14-3-3ζ s dvojitou mutací, kteří projevují kompletní monomerizaci v širokém rozpětí koncentrací a přitom si zachovají charakteristickou alfa šroubovicovou strukturu. Monomerní charakter navržených mutantů byl potvrzen analytickou centrifugací a maloúhlým rentgenovým rozptylem. Diferenciální skenovací kalorimetrie ukázala, že stabilita monomerních mutantů je nižší než 14-3-3ζ divokého typu. Krystaly získané krystalizací 14-3-3ζ monomerních mutantů neposkytly rozumné difrakční vzorce nebo dostatečné rozlišení pro vyřešení struktury. Na druhou stranu nukleární magnetická resonance přinesla částečný náhled do jejich struktury. NMR HN-TROSY experimenty odhalily, že monomerní mutanti poskytují velmi podobné NMR spektra jako 14-3-3ζ divoký typ. Poněvadž dimerní struktura 14-3-3ζ divokého typu je dobře popsána pomocí několika krystalografických struktur, můžeme předpokládat, že se monomerní mutanty mají podobnou terciární strukturu.

Abstract:

The dimeric character of 14-3-3 proteins is often crucial for their functions and for their ability to bind phosphorylated binding partners. 14-3-3s form homo/heterodimers that are in dynamic equilibrium with monomers. The dimeric form of 14-3-3 proteins has been heavily studied, however, structure and properties of the monomeric form remain insufficiently described. The monomerization of 14-3-3ζ is regulated in different ways, including phosphorylation of Ser58 located at the dimer interface. Phospho-mimicking mutant (S58E) of 14-3-3ζ causes only partial dissociation. However, we designed two mutants of 14-3-3ζ with a double mutation that exhibit complete monomerization over a wide range of concentrations while retaining the characteristic α-helical structure based on our far-UV CD measurements. The monomeric character of designed mutants was proved by analytical ultracentrifugation and small angle X ray scattering. Differential scanning calorimetry showed that the stability of monomeric mutants is lower than 14-3-3ζ WT. The crystals obtained by the crystallization of 14-3-3ζ monomeric mutants did not provide reasonable diffraction pattern or sufficient resolution for structure solving. On the other hand, nuclear magnetic resonance provided partial insight into the structure. NMR HN-TROSY experiments revealed that the monomeric mutants possess very similar overall NMR fingerprints as 14-3-3ζ WT. Since the dimeric structure of the 14-3-3ζ WT is well described by several X-ray structures, we can assume that monomeric mutants possess a similar tertiary structure.
