Analysis of chromatin SMC5/6 complex subunits

Abstract:

Komplexy SMC jsou důležitou součástí organizace a architektury chromozomů v průběhu buněčného cyklu. Proteinový komplex SMC5/6 (Structural Maintenance of Chromosomes) se řadí mezi evolučně konzervované eukaryotické SMC komplexy. Hraje důležitou roli při opravách DNA, stabilizaci replikačních vidlic a také se podílí na alternativním prodlužování telomer a inhibici transkripce virů. Tento komplex je tvořen heterodimerem základních SMC proteinů – SMC5 a SMC6. Zároveň obsahuje podjednotky NSE1-6. Nejméně prozkoumána je podjednotka NSE5. Napříč eukaryotickými organismy jsou známé pouze interakce s podjednotkou NSE6, ale její interakce s ostatními proteiny komplexu jsou neznámé. Proto se tato práce zaměřuje na analýzu protein-proteinových interakcí podjednotky NSE5 u modelového organismu Physcomitrium patens (Pp) v komplexu SMC5/6. Za použití metod kvasinkového dvouhybridního systému (Y2H) a pull down byly charakterizovány interakce s proteiny PpNSE6, PpSMC5 a PpSMC6. Bylo zjištěno, že C-koncová část podjednotky PpNSE5 interaguje s podjednotkou PpNSE6, zatímco N-koncová oblast interaguje s PpSMC6 a PpSMC5, z níž je pro interakci s proteinem PpSMC6 kritických prvních 71 amk. Zároveň se podjednotka PpNSE5 váže na protein PpSMC5 v oblasti pantové domény, avšak na protein PpSMC6 se váže blíže hlavové doméně. Na základě této analýzy byly detailněji zmapovány interakce PpNSE5 s vybranými podjednotkami komplexu SMC5/6. Výsledky této práce by mohly být oporou pro budoucí studium PpSMC5/6 komplexu.

Abstract:

SMC complexes are an important components of chromosome organization and architecture during the cell cycle. The SMC5/6 (Structural Maintenance of Chromosomes) protein complex is one of the evolutionarily conserved eukaryotic SMC complexes. It plays an important role in DNA repair, stabilization of replication forks and is also involved in alternative telomere elongation and inhibition of viral transcription. The SMC5/6 complex consists of a heterodimer of the basis SMC proteins - SMC5 and SMC6. It also contains NSE1-6 subunits. The NSE5 subunit is the least studied. Across eukaryotic organisms, only interactions with the NSE6 subunit are known, but its interactions with other proteins of the complex are unknown. Therefore, this work focuses on analyzing the protein-protein interactions of the NSE5 subunit in the model organism Physcomitrium patens (Pp) within the SMC5/6 complex. The interactions with PpNSE6, PpSMC5 and PpSMC6 proteins were characterized using yeast two-hybrid (Y2H) and pull-down methods. It was found that the C-terminal region of the PpNSE5 subunit interacts with the PpNSE6 subunit, whereas the N-terminal region interacts with PpSMC6 and PpSMC5. Of which, the first 71 amino acids are critical for interaction with the PpSMC6 protein. Furthermore, the PpNSE5 subunit binds to the PpSMC5 protein in the hinge domain region but binds to the PpSMC6 protein closer to the head domain. Based on this analysis, the interactions of PpNSE5 with selected subunits of the SMC5/6 complex were mapped in more detail. The results of this work could underpin future studies of the PpSMC5/6 complex.
