Analysis of stool microbiome composition of child from birth to 1 year of life

Abstract:

This bachelor thesis deals with the analysis of the intestinal microbiome composition of one child from birth to the completion of hers first year. The theoretical part of the thesis contains basic description of human microbiome and factors that influence its composition. In the following part I describe the compositional data, their transformations and statistical tests to compare two selections, which are later used in the practical part. Thereinafter it is described the bacterial data collection and analysis, their basic characteristics and bacterial distribution into homogeneous periods in order to find breakpoints when the change in the microbial composition occurs. Output of this thesis depicts what factor influenced the microbiome of the child in his first year. Diet, probiotic use, and changing the environment have been shown to be factors that affect microbes, compared with vaccination, after which no changes have been detected in the microbiome.

Abstract:

Tato bakalářská práce se zaobírá analýzou složení střevního mikrobiomu jednoho dítěte od jeho narození do dovršení jeho prvního roku. Teoretická část obsahuje základní informace o lidském mikrobiomu, ovlivňujících faktorech a jejich dopadu na mikrobiom. V další části jsou popsána kompoziční data, jejich transformace a statistické testy pro porovnání dvou výběrů, které jsou později využity v praktické části. Dále je popsán sběr vzorků a analýza dat bakteriálního složení, jejich základní charakteristika a rozdělení do homogenních období za účelem nalezení časových zlomů, kdy nastala v mikrobiomu změna. Výstupem práce je seznam faktorů, které ovlivňují mikrobiom dítěte v jeho prvním roce života. Patří sem strava, užívání probiotik a změna prostředí dítěte, na rozdíl od očkování, po kterém v mikrobiomu žádné změny zachyceny nebyly.
