Honeybee as a biomonitor of gold occurrence in a locality

Abstract:

Tato diplomová práce se zabývá možností využití včely medonosné (Apis mellifera) jako pasivního biomonitoru pro detekci zlata v životním prostředí. Včely, díky svému rozsáhlému kontaktu s vegetací a povrchem krajiny, mohou sloužit jako indikátor přítomnosti anorganických částic, včetně ušlechtilých kovů. Cílem práce bylo ověřit, zda lze obsah Au ve včelím těle stanovit rutinním analytickým postupem a zda takto získaná data mohou diferencovat mezi referenčními a zlatonosnými lokalitami. V rešeršní části byla zmapována historie těžby zlata v českých zemích, přehled ložisek a současné průzkumné metody. Byla diskutována ekologická zátěž spojená s těžbou i potenciál méně invazivních průzkumných technik. Pro analýzu Au byla optimalizována metodika zahrnující mikrovlnný ohřev (MW) a následnou koncentraci extraktu na topné desce (HP), s využitím Leffortovy lučavky jako extrakčního činidla. Kvantifikace Au byla provedena pomocí hmotnostní spektrometrie s indukčně vázaným plazmatem (ICP-MS), přičemž byl dosažen detekční limit 0,36 ng/g. V práci byly rovněž identifikovány a eliminovány spektrální interference (např. TaO⁺, HfOH⁺), které mohou ovlivnit přesnost stanovení ¹⁹⁷Au. Výsledky ukázaly, že metoda umožňuje detekci Au ve stopových koncentracích. Zatímco ve vzorcích z referenčních lokalit nebylo Au detekováno (s výjimkou jednoho případu), ze všech čtyř zlatonosných oblastí byly získány hodnoty nad mezí detekce metody (LOD). Práce tak potvrzuje, že stanovení Au ve včelím těle může být efektivním a ekologicky šetrným nástrojem pro předběžný geochemický screening. V kombinaci s dalšími podpůrnými metodami může tato strategie rozšířit možnosti nerušícího průzkumu v zájmových oblastech.

Abstract:

This thesis explores the potential of the honey bee (Apis mellifera) as a passive biomonitor for detecting gold (Au) in the environment. Due to their extensive contact with vegetation and surface structures, honey bees can serve as indicators of the presence of inorganic particles, including noble metals. The aim of the study was to verify whether the gold content in bee tissue can be determined using a routine analytical method and whether the resulting data can be used to differentiate between reference and gold-bearing sites. The literature review provides an overview of the history of gold mining in the Czech lands, a summary of known deposits, and current exploration techniques. The environmental burden associated with gold mining is discussed, along with the potential of less invasive exploration approaches. For the analysis of Au, a method was optimized that combines microwave-assisted digestion (MW) with subsequent solvent evaporation on a heating plate (HP), using Leffort’s aqua regia as the extraction medium. Gold quantification was performed using inductively coupled plasma mass spectrometry (ICP-MS), achieving a method detection limit (LOD) of 0.36 ng/g. Spectral interferences (e.g., TaO⁺, HfOH⁺), which can affect the accuracy of ^197Au determination, were identified and effectively minimized. The results demonstrated that the method enables the detection of Au at trace levels. While no Au was detected in samples from reference sites (with the exception of one case), measurable values above the LOD were obtained in all four gold-bearing regions. These findings confirm that determining Au content in bee bodies can serve as an effective and environmentally friendly tool for preliminary geochemical screening. When used alongside complementary methods, this approach can enhance the potential for non-invasive mineral exploration in areas of interest.
