Application of lab-on-a-chip technology in the treatment of cancer

Abstract:

Tato práce je zaměřena na aplikaci lab-on-a-chip technologie v léčbě nádorových onemocnění. Blíže popsaným onemocněním je chronická B-lymfocytární leukémie. Teoretická část se zabývá léčbou hematoonkologických malignit a s ní často spojenou rezistenci k lékům. Dále se práce zabývá apoptózou a vybranými metodami k jejímu průkazu, jelikož cílem chemoterapeutické léčby je navození apoptózy maligních buněk. V poslední kapitole teoritické části je popsána distribuovaná dielektroforetická cytometrie (2DEP cytometrie), lab-on-a-chip technologie, jež byla použita v experimentální části. Praktická část se věnuje měření B-lymfocytů izolovaných z krve pacientů s prokázanou chronickou B-lymfocytární leukemií a jejich reakci na léčbu. Účinnost léčby byla v podobě sledování progrese apoptózy monitorována 2DEP cytometrií. Výsledky naměřené pomocí 2DEP cytometrie byly porovnávány s konvenčními metodami (Alamar blue, 7-Aminoaktinomycinem D a barvením trypanovou modří). Výsledky práce poukazují na potenciál 2DEP cytometrie konkurovat konvenčním metodám pro měření buněk, jejich vlastnosti a identifikaci důležitých biomarkerů, a to i bez nutnosti barvení či jiného značení buněk.

Abstract:

This thesis is focused on the application of lab-on-a-chip technology in the treatment of cancer. The disease described herein is chronic B-cell leukemia. The theoretical part deals with the treatment of haemato-oncological malignancies and the corresponding drug re-sistance that is often associated with them. Furthermore, the thesis deals with apoptosis and with selected methods for its detection, as the goal of chemotherapeutic treatment is to induce apoptosis of malignant cells. The last chapter of the theoretical part describes dis-tributed dielectrophoretic cytometry (2DEP cytometry), lab-on-a-chip technology, which is used in the experimental part. The practical part deals with the measurement of B-lymphocytes isolated from the blood of patients with confirmed chronic B-cell leukemia and their response to treatment. Treatment efficiency was monitored by 2DEP cytometry and corresponds to the progression of apoptosis. 2DEP cytometry results were compared with conventional methods (Alamar blue, 7-aminoactinomycin D and trypan blue dying). The results of the work show the potential of 2DEP cytometry to compete with conven-tional methods for measuring cells, their properties and identifying important biomarkers, even without dying or other labeling of the cells.
