Extraction of nitroderivatives of toluene and their degradation products from soil

Abstract:

Vzhledem k celosvětovému rozšíření a toxikologickým vlastnostem nitroderivátů toluenu a metabolitů jejich degradace, je velmi důležité vyvinout přesnou metodu izolace a detekce těchto sloučenin z environmentálních matric. Předkládaná diplomová práce srovnává stávající metody extrakce výbušnin z půdy. Byla vyvinuta metoda detekce souboru výbušnin a jejich degradačních produktů definovaných v U.S. EPA Method 8330. K separaci byla použita vysoce účinná kapalinová chromatografie s reverzním uspořádáním fází s UV-detekcí (RP-HPLC-UV) doporučená U.S. EPA. Práce srovnává čtyři extrakční techniky – extrakci pomocí automatickém extraktoru Soxhletova typu, tlakovou extrakci rozpouštědlem (PSE), mikrovlnnou extrakci (MAE) a superkritickou fluidní extrakci (SFE). Byly prověřeny základní extrakční podmínky – použití extrakčního rozpouštědla, teplota, tlak a doba extrakce. Ze srovnání výsledků extrakčních metod plyne, že nejvhodnější metodou izolace této sady sloučenin je tlaková extrakce rozpouštědlem, u které se výtěžnosti pohybovaly v rozsahu 65-112%. Následuje extrakce na automatickém extraktoru Soxhletova typu, kde bylo dosaženo výtěžnosti 65-99%, a superkritická fluidní extrakce (52-75%). Nejméně vhodnou metodou je mikrovlnná extrakce, kde se výtěžnosti pohybují v rozmezí od 28 do 65%. Ve všech případech byl z půdy obtížně izolován tetryl. Nejlepších výsledků bylo u této látky dosaženo za použití tlakové extrakce rozpouštědlem (58%). Srovnání extrakce analytů z čerstvě kontaminované půdy a vzorků, které prošly procesem stárnutí, je v této práci zahrnuto také. Použitou extrakční technikou byla Soxhletova extrakce. Z 6-týdenního experimentu plyne, že proces stárnutí ovlivňuje výtěžnosti extrakcí všech sledovaných látek. U některých látek došlo v průběhu experimentu k postupnému poklesu výtěžností (2,4,6-trinitrotoluen, 1,3,5-trinitrobenzen, 1,3-dinitrobenzen, 4-amino-2,6-dinitrotoluen a 3-nitrotoluen), u 2,6-dinitrotoluenu a 2,4-dinitrotoluenu množství extrahované z půdy vzrostlo.

Abstract:

Considering worldwide spread and toxicological properties of nitroderivatives of toluene and metabolites of their degradation, it is very important to develop an accurate extraction and detection method of these substances from environmental matrices. The present diploma thesis compares current methods of extraction of explosives from soil. Detection method for determination of explosive compounds defined by U.S. EPA Method 8330 was modified. Reversed-phase high performance liquid chromatography equipped with UV-detection (RP-HPLC-UV) was used for separation of these substances. This paper compares four extraction techniques – extraction using automated extractor of Soxhlet type, pressurized solvent extraction (PSE), microwave assisted extraction (MAE) and supercritical fluid extraction (SFE). Basic extraction conditions were investigated – use of extraction solvent, temperature, pressure and extraction time. When comparing optimized extraction methods, the most suitable isolation method of these compounds from soil is pressurized solvent extraction. Recoveries of this technique were in the range from 65 to 112%. Extraction recoveries of Soxhlet warm extraction reached 65-99% and supercritical fluid extraction 52-75%. The lowest extraction recoveries (28-65%) were obtained using microwave assisted extraction. Tetryl was difficultly extracted from soil in all cases. The best results were reached by pressurized solvent extraction (58%). Extraction of analytes from freshly spiked soil and from soil that went through ageing process are compared in this paper. Extractions were performed using Soxhlet warm extractor. Experiment lasted 6 weeks. Results show that ageing process influences extraction recoveries of all monitored compounds. Recoveries of some substances decreased during the experiment (2,4,6-trinitrotoluene, 1,3,5-trinitrobenzene, 1,3-dinitrobenzene, 4-amino-2,6-dinitrotoluene and 3-nitrotoluene). Recoveries of 2,6-dinitrotoluene and 2,4-dinitrotoluene increased.
