Purification and analysis of microbial protins using chromatographic and electrophoretic methods

Abstract:

Nukleokapsidové proteiny N evropského a severoamerického sérotypu viru repro¬dukčního a respiračního syndromu prasat (PRRSV) byly připraveny v eukaryotním bakulovi¬rovém systému genové exprese. Produkce a identita obou rekombinantních nukleokapsido¬vých N proteinů byly prokázány pomocí SDS-PAGE. Specifita rekombinantních proteinů v rekombinantních bakulovirech byla ověřena imunoblotem pomocí anti PRRS prasečího séra a monoklonální protilátky proti viru PRRS. Exprimované proteiny N obou sérotypů byly izolovány z lyzátu hmyzích buněk pomocí preparativní elektro-eluční SDS-PAGE na zařízení Prep Cell Model 491 (Bio-Rad) ve spojení se systémem FPLC (Pharmacia). Koncentrace purifikovaných proteinů N v získaných frakcích, které byly následně analyzová¬ny pomocí SDS-PAGE a imunoblotu, dosahovala hodnot v rozmezí 80 - 250 µg/ml. Celkový výtěžek dosahoval 1 mg během jednoho běhu purifikace při zachování čistoty přes 95%. Frakce obsahující čistý nukleokapsidový protein N byly sloučeny a použity jako antigeny k sestavení diagnostického nepřímého ELISA testu k průkazu PRRSV protilátek v sérech prasat. Porovnáním tohoto ELISA testu se standardní diagnostickou soupravou IDEXX HerdCheck ELISA na souboru 552 sér prasat byla stanovena citlivost 98.3 % a specifita 92.6 %. Z anaerobně kultivovaného kmene Pd2121 bakterie Paracoccus denitrificans defektní¬ho v cytochromu c550 byl purifikován kombinací iontoměničové a gelové permeační chromatografie periplasmatický protein schopný přenášet elektrony z cytoplasmatické membrány na periplasmatickou nitritreduktázu. Pomocí SDS-PAGE byla potvrzena čistota izolovaného proteinu a stanovena molekulová hmotnost 14 kDa. Použitím analytické gelo¬vé permeační chromatografie byla zjištěna stejná velikost a zároveň prokázán výskyt tohoto přenašeče v nativním stavu jako monomeru. Několika dalšími nezávislými charakteristikami (obsah mědi, spektrum ve viditelné oblasti, N-terminální aminokyselinová sekvence) bylo dokázáno, že purifikovaným proteinem je pseudoazurin. Dva rozpustné cytoplasmatické enzymy (FerA a FerB) bakterie Paracoccus denitri¬ficans s Fe(III) reduktázovou aktivitou byly purifikovány kombinací aniontoměničové chromatografie (Sepharose Q), chromatofokusace (Mono P) a gelové permeační chromatogra¬fie (Superose 12). Tyto dva enzymy byly dále charakterizovány celou řadou biochemických metod. Byly stanoveny molekulová hmotnost, v případě jednoho z nich potvrzena oligomerní struktura, isoelektrický bod, koenzymové složení, N-terminální aminokyselinové sekvence, substrátové specifity a byly provedeny kinetické studie. FerA je monomer o molekulové hmotnosti 19 kDa, zatímco nativní FerB vykazuje velikost okolo 50 kDa a je tvořen ze dvou stejných podjednotek o velikosti 20 kDa. FerA může být klasifikován jako NADH:flavin oxidoreduktáza se sekvenčním reakčním mechanismem a vyžaduje pro svoji Fe(III) reduktá¬zovou aktivitu přítomnost flavinu jako disociabilního kosubstrátu. Purifikovaný FerB obsahu¬je nekovalentně vázaný redoxně aktivní koenzym FAD a může využít NADPH místo NADH, neredukuje volný flavin a vykazuje ping-pongový kinetický model se substráty NADH a Fe(III)-NTA. Byly určeny N-koncové aminokyselinové sekvence FerA a FerB. V případě FerB jsme zjistili vysokou podobnost s chromanreduktázou Pseudomonas putida, která byla potvrzena schopností redukovat chroman za přítomnosti NADH. N-koncová sekvence enzy¬mu FerA nejeví významnou podobnost s žádnou známou oxidoreduktázou. Byla provedena proteinová analýza hyfového a sporového antigenu patogenní houby Trichofyton verrucosum pomocí SDS-PAGE. Imunoblotem byla charakterizo¬vána molekulová hmotnost proteinů, které jsou zodpovědné za jejich specifickou imunitní reakci. Oba antigeny AgH a AgS vykazovaly stejn

é dominantní proteiny, nicméně ale drobné rozdíly v jejich proteinovém spektru mohou být příčinou relativně významných odlišností v imunitních reakcích a to jak humorálních, tak i buněčných.

Abstract:

The nucleocapsid proteins N of European and North American serotypes of porcine reproductive and respiratory syndrome virus (PRRSV) were constructed in the eukaryotic baculovirus system of gene expression. Production and identity of both nucleocapsid proteins N were proven using SDS-PAGE. Specificity of recombinant proteins in recombinant baculoviruses was tested by immunoblot technique using anti-PRRS serum from swine and monoclonal antibodies against PRRS virus. The expressed proteins N of both serotypes were isolated from the insect cell lysate by the preparative SDS-PAGE and electroelution using the Prep Cell Model 491 (Bio-Rad) apparatus connected to the system FPLC (Pharmacia). Concentrations of purified proteins N in the obtained fraction, which were subsequently detected by means of SDS-PAGE and immunoblot analysis, ranged between 80 - 250 µg/ml. The total yield for one purification run reached 1 mg with purity being more than 95%. Fractions containing pure nucleocapsid protein N were combined and used as antigens for production of an indirect ELISA test for detection of anti-PRRSV antibodies in serum from pigs. This ELISA test was compared with a standard diagnostic kit IDEXX HerdCheck® ELISA using a group of sera from 552 pigs; sensitivity and specificity were found to be 98.3 % and 92.6 %, respectively. Periplasmic protein able to transfer electrons from cytoplasmic membrane to the periplasmic nitrite reductase has been purified from the anoxically grown cytochrome c550 –deficient strain Pd2121 of bacteria Paracoccus denitrificans by combination of ion-exchange and gel permeation chromatography. Purity of the isolated protein was confirmed by means of SDS-PAGE and molecular mass was shown to be 14 kDa. The same size was detected by analytical gel permeation chromatography. It was found that this transmitter is monomer in native state. The purified protein was shown to be pseudoazurin by several independent criteria (copper content, visible spectrum, N-terminal amino acid sequence). Two soluble cytoplasmic enzymes (FerA and FerB) from bacterium Paracoccus denitrificans with Fe(III) reductase activity were purified by combination of anion-exchange chromatography (Sepharose Q), chromatofocusing (Mono P) and gel permeation chromatography (Superose 12). These two enzymes were further characterised by a number of biochemical methods. Molecular mass (quaternary structure was confirmed in one of them), isoelectrical point, content of coenzymes, N-terminal amino acid sequence and substrate specificity were assessed and kinetic studies were performed. FerA is a monomer with molecular mass of 19 kDa, whereas native FerB exhibits molecular mass of about 50 kDa and consists of two identical subunits of about 20 kDa. FerA can be classified as NADH:flavin oxidoreductase with a sequential reaction mechanism. It requires presence of flavine as dissociable co-substrate for reductase activity on Fe(III). The purified FerB contains a noncovalently bound redox-active FAD coenzyme and it can utilise NADPH instead of NADH, does not act to reduce free flavine and gives a ping-pong type of kinetic pattern with substrates NADH and Fe(III)-NTA. N-terminal amino acid sequences FerA and FerB were detected. In the case of FerB, we found high similarity with chromate reductase from Pseudomonas putida, which was confirmed by the ability to mediate chromate reduction in the presence of NADH. The N-terminal sequence of enzyme FerA did not bear a significant homology to any known oxidoreductase. Protein analysis of antigens containing hyphae and spores in pathogenic fungus Trichofyton verrucosum was performed by means of SDS-PAGE. Molecular mass of proteins was characterised by immunoblot analysis; these proteins are responsible for the specific immune response. The response of the same dominant proteins was observed for both antigens AgH and AgS; nevertheless small dissimilarities in their protein spectra could cause quite important differences
