Study of recombinant hyaluronidase TsHr in the context of low molecular weight hyaluronic acid degradation

Anotácia:

Hyaluronidázy jsou významnou skupinou enzymů schopných katalyzovat depolymeraci kyseliny hyaluronové. Výzkumy posledních let se zaměřují na identifikaci nových hyaluronidáz a na objasnění jejich mechanismu degradace. Tato práce se zabývá studiem nové rekombinantní hyaluronidázy z Talaromyces stipitatus (TsHr) a to z pohledu degradace krátkých hyaluronanových oligosacharidů. Testované oligosacharidy se lišily svojí délkou a modifikací na redukujícím nebo neredukujícím konci. Průběh enzymatické reakce byl monitorován ve velmi krátkých časových intervalech kolorimetrickou metodou podle Morgana a Elsona a kapalinovou chromatografií spojenou s hmotnostní detekcí. Získaná data kinetických parametrů TsHr potvrdila vliv délky oligosacharidu na tyto parametry. Nejvyšších počátečních rychlostí bylo dosaženo v případě degradace substrátu s 22 podjednotkami, naopak k degradaci tetrasacharidu za testovaných podmínek nedocházelo. Modifikace redukujícího konce hexasacharidu měla negativní dopad na počáteční rychlosti degradace. S rostoucí délkou oligosacharidu tento vliv klesal. Naopak modifikace neredukujícího konce spíše aktivitu TsHr podpořilo. Rovněž byla v rámci experimentální části práce potvrzena nízká citlivost kolorimetrické metody a její nespolehlivost pro monitorování kinetiky enzymatické reakce.

Abstract:

Hyaluronidases are an important group of enzymes capable of catalyzing the depolymerization of hyaluronic acid. Recent research has focused on identifying new hyaluronidases and clarification their mechanism of degradation. This work deals with the study of a new recombinant hyaluronidase from Talaromyces stipitatus (TsHr) in terms of short hyaluronan oligosaccharides degradation. Tested oligosaccharides differed in length and modification at the reducing or non-reducing. The progress of the enzymatic reaction was monitored at very short time intervals by the Morgan and Elson colorimetric method and by liquid chromatography with mass detection. The obtained data of kinetic parameters TsHr confirmed the effect of oligosaccharide length on these parameters. The highest initial rates were reached in the case of degradation of the substrate with 22 subunits, on the contrary, the degradation of tetrasaccharide did not occur under the tested conditions. Modification of the reducing end of the hexasaccharide had a negative impact on the initial degradation rates. This effect decreased with increasing oligosaccharide length. In contrast, modification of the non-reducing end promoted TsHr activity. The low sensitivity of the colorimetric method and its unreliability for monitoring the kinetics of the enzymatic reaction were also confirmed in the experimental part of the work.
