Bioinformatic searching for and comparison of lectins from Photorhabdus

Abstract:

Lektiny jsou proteiny neimunitního původu, které mají vlastnost specificky a reverzibilně vázat sacharidy. Lektiny se objevují téměř ve všech živých organismech, kde zastávají velké množství funkcí. Živočišné lektiny jsou převážně součástí imunitního systému a pomáhají buňkám imunitního systému v navádění do místa zánětu a infekce. Bakteriální lektiny jsou často exprimovány na povrchu buněk a mohou se účastnit adhezivních reakcí, které vedou k napadnutí, infikování a následné smrti oběti. V této bakalářské práci se věnujeme lektinům z bakterií Photorhabdus, které žijí v symbiotickém vztahu s hlísticemi rodu Heterorhabditis. Hlístice žijí v půdě, kde aktivně vyhledávají hmyzí oběť, kterou s pomocí bakteriálního symbionta usmrcují. Na pracovišti výzkumné skupiny Glykobiochemie bylo identifikováno 5 lektinů z bakterie P. luminescens subsp. laumondii TT01. Tato práce se zabývá bioinformatickým vyhledáváním a srovnáním těchto lektinů s homologními proteiny Photorhabdus v databázích a konstrukcí fylogenetických stromů z jejich aminokyselinových sekvencí. Součásti práce je i diskuze o zdůvodnění počtu genů kódující lektiny Photorhabdus.

Abstract:

Lectins are proteins of a nonimmune origin able to bind sacharides reversibly and in a very specific way. They occur across all living organisms, where they occupy a lot of biological funcions. Animal lectins are usually part of the immune system and helping the immune cells in trafficing into the site of inflammation and infection. Bacterial lectins are expressed on the surface of microorganisms, playing part in adhesive reactions. This leads to attacking, infection and death of a host. In this theses we are dealing with lectins from Photorhabdus bacteria which are surviving in a symbiotic relationship with nematodes of Heterohabditis genus. Heterohabditis nematodes live in a soil, where they are searching for a posibble insect victim, then the insect is killed by their symbiotic bacteria. On Glycobiochemistry group workplace 5 lectins from Photorhabdus luminescens subsp. laumondii TT01 were identified. This thesis is focused on bioinformatic search and comparison of these lectins and their homologous proteins from Photorhabdus genus, which can be found in databases and construction of phylogenetic trees from their amino acid sequences. Part of this theses is also focused on discussion about the number of genes which are coding lectins from Photorhabdus.
