Evaluation of the use test with Vibrio fischeri for real samples of the contaminated environment

Abstract:

Tématem diplomové práce jsou testy toxicity využívající jako modelový organismus bakterii Vibrio fischeri. Mezi tyto testy patří LUMIStox, MICROTOX, ToxAlert a Deltatox, test s Vibrio fisheri, který probíhá v kinetickém režimu Biotox flash test, test pracující s pevnými vzorky Microtox solid phase a test genotoxicity Mutatox. Hlavním cílem teoretické části bylo zhodnocení těchto testů při měření s reálnými vzorky, u kterých došlo k průmyslové kontaminaci. V práci jsou popsány stručné metodiky těchto testů, základní informace a praktické využití. Praktická část práce se zabývá zjištěním toxického efektu u reálných vzorků pomoci testu LUMIStox. Jako testované vzorky byly zvoleny otěry z brzdového obložení od různých výrobců. Cílem bylo zjistit akutní toxicitu při využití Vibrio fischeri jako modelového organismu. Jako nejtoxičtější vyšel vzorek č. 2, kdy se jednalo o čisté fenol-formaldehydové pryskyřice, které slouží v brzdovém obložení jako pojiva.

Abstract:

The toxicity tests are the subject of this thesis. They use the bacterium Vibrio fischeri as a model organism. These tests include LUMIStox, MICROTOX, ToxAlert and Deltatox, test with Vibrio fischeri which proceeds the kinetic mode of Biotox flash test, test working with solid samples Microtox solid phase, and test of genotoxicity Mutatox. The work deals also with the evaluation of these tests using measurements with real samples affected by industrial contamination. It describes their brief procedures, basic informations and their utilization. The practical part investigates the findings of toxic effects at real samples with help the LUMIStox test. The samples were created by the abrasions from the brake pads from various producers. The aim is to detect an acute toxicity with the use of Vibrio fischeri as a model organism. According to the measurment, the used sample 2 was the most toxic, the sample is represented by phenol formaldehyde resins used as a binder in brake pads.
