Komparativní genomika Treponema pallidum: cesta od určení sekvence chromozomu k aplikacím v klinické diagnostice

Comparative genomics of Treponema pallidum: from whole genome sequencing to diagnostic applications
Abstract:
Druh Treponema pallidum zahrnuje 3 pro člověka patogenní poddruhy. Tyto poddruhy – pallidum, pertenue a endemicum – způsobují přes velice blízkou sekvenční příbuznost onemocnění s odlišnou klinickou manifestací. Jedná se o syfilis, yaws a endemickou syfilis. Patogenní treponemata zahrnují ovšem i patogeny jiných hostitelů než člověka. Jedná se o druhově nezařazený opičí izolát Fribourg-Blanc a druh …more
Abstract:
h pro poddruh pallidum, pertenue a endemicum. Konsenzuální nukleotidové sekvence čtyř lokusů byly použity pro rekonstrukci fylogenetického stromu. Fylogenetická analýza potvrdila opodstatnění rozdělení patogenních treponemat na jednotlivé druhy a poddruhy. Nepřinesla ovšem zásadní vhled do dlouhodobé debaty o původu venerické syfilis. Srovnání genomů kmenů Nichols a SS14 odhalilo sekvenčně divergentní …more
Abstract:
Genus Treponema includes several pathogenic spirochetes. Three human pathogens include: Treponema pallidum ssp. pallidum, the causative agent of venereal syphilis; T. pallidum ssp. pertenue, the causative agent of yaws; and T. pallidum ssp. endemicum causing endemic syphilis. Non-human pathogens include: unclassified simian isolate Fribourg-Blanc and T. paraluiscuniculi. Fribourg-Blanc strain causes …more
Abstract:
led divergent loci that can be used for molecular typing of syphilitic clinical isolates. As T. pallidum can not be cultured in vitro, molecular detection of syphilitic treponemes represents the only available method for treponeme typing. In addition, macrolide antibiotic resistance in clinical isolates can be tested. Our aim was to screen divergent loci of the T. pallidum genomes and to assess the …more
