Preparation and characterization of nanoparticle bioconjugates

Abstract:

Katalytické nanočástice představují nový způsob značení biomolekul pro imunochemické detekční metody, který v budoucnu může nahradit nákladné, méně katalyticky aktivní a méně stabilní enzymy. Teoretická část této diplomové práce se zabývá využitím katalytických nanočástic v bioanalytických metodách. Experimentální část je zaměřena na přípravu nanočástic pruské modři, které v přítomnosti H2O2 katalyzují oxidaci 3,3',5,5'-tetramethylbenzidinu za vzniku snadno detekovatelných barvených produktů. Bylo zjištěno, že reakční kinetiku katalyzované reakce lze popsat pomocí modelu Michaelis-Mentenové. Číslo přeměny (~104 s-1) pro tuto reakci bylo řádově vyšší než číslo přeměny dříve popsaných nanočástic z materiálu Fe3O4 nebo Co3O4. Nanočástice pruské modři byly biotinylovány a použity pro imunochemickou detekci lidského sérového albuminu s limitem detekce 0,26 µg.ml-1 a rozsahem měření od 0,34 do 1,29 µg.ml-1. Připravené nanomateriály byly charakterizovány pomocí gelové elektroforézy, mikroskopie atomových sil, měření dynamického rozptylu světla a transmisní elektronové mikroskopie.

Abstract:

Catalytic nanoparticles represent a new method for the labeling of biomolecules for immunochemical detections, which may replace cost, less catalytically active and less stable enzymes. The theoretical part of this thesis describes the use of catalytic nanoparticles in bioanalytical methods. The experimental part is focused on the prepara-tion of Prussian blue nanoparticles, which catalyze the oxidation of 3,3',5,5'-tetramethylbenzidine in the presence of H2O2 to form a readily detectable colored prod-ucts. It was found that the reaction kinetics of the catalyzed reaction may be described by the model of Michaelis-Menten. The turnover number (~104 s-1) for this reaction was much higher than a turnover number of previously described nanoparticles composed of Fe3O4 or Co3O4. Prussian blue nanoparticles were biotinylated and used for the immunochemical detection of human serum albumin with the limit of detection 0,26 μg.ml-1 and working range from 0,34 to 1,29 μg.ml-1. Prepared nanomaterials were characterized by gel electrophoresis, atomic force microscopy, dynamic light scattering and transmission electron microscopy.
