The antibody production against the selected defence proteins in tobaco and wine plants

Abstract:

Tato diplomová práce se zabývala přípravou rekombinatních obranných proteinů chitinázy (PR3) a glukanázy (PR2), které jsou součásti obranné reakce u rostlin. Vytvořené proteiny budou základem k výrobě protilátek. Jejich využití bude k dalšímu zkoumání obranného mechanismu u rostlin. Nejprve byla vyizolována RNA z listu tabáku 10 h po aplikaci elicitoru cryptogeinu. Následně byla provedena reverzní transkripce vyizolované RNA a amplifikace genů pro oba proteiny PR2 i PR3. Během amplifikace byly do genů zavedeny restrikční místa pro endonukleázy NcoI a XhoI pro snadné vloklonování do expresního vektoru pET-32b. Exprese proteinů probíhala v buňkách E. Coli ORIGAMI DE3, které mají mutaci v genech pro thioredoxin reduktázu. Tato mutace velmi zvyšuje tvorbu disulfidických můstků v cytoplazmě, což podporuje správné sbalení proteinu. Po transformaci do buněk ORIGAMI DE3 byla provedena testovací exprese na základě, které byly vybrány správné podmínky pro finální expresi proteinu ve velkém množství. Sklizené buňky byly lyzovány, získaný lyzát byl přečištěn na IMAC (afinitní chromatografie s imobilizovaným iontem kovu). Získané frakce byly podrobeny dialýze a následně SDS-Page a Western-blot analýze pro potvrzení přítomnosti proteinu. Proteiny PR2 a PR3 se v první fázi nepodařilo vyprodukovat. Byla provedena nová exprese proteinu PR3, kdy byla z genu odstraněna sekvence signálního (sekrečního) peptidu. Po odstranění signální sekvence byla exprese proteinu úspěšná. Signální peptid měl pravděpodobně negativní vliv na správné sbalení proteinu, který pak přecházel do inkluzních tělísek. Stejný postup odstranění signální sekvence je třeba tedy provést i při produkci proteinu PR2.

Abstract:

Diploma thesis is focused in the preparation of recombinant proteins defensive chitinase (PR3) and glucanase (PR2), which are components of defense responses in plants. Created proteins will be the basis for the production of antibodies. Their use will be to further investigating of defense mechanism in plants. RNA was purified from tobacco leaves 10 hours after aplication of elicitor cryptogein. RNA was put to reverse transcription. From obtained DNA the genes for PR2 and PR3 were amplified by PCR . During the amplification the restriction sites for endonucleases NcoI, XhoI were inerted in to the gene for easy cloning into the expression vector PET-32b. Protein expression was made in E. coli ORIGAMI DE3 cells, which have a mutation in the gene for thioredoxin reductase. This mutation greatly increases the production of disulfide bridges in the cytoplasm, which promotes proper protein packing. After transformation into cells ORIGAMI DE3, expression testing was performed on the basis that the right conditions were selected for the final expression of the protein in large quantities. The harvested cells were lyzed, obtained lysates were purified on IMAC (affinity chromatography with immobilized metal ion). The obtained fractions were subjected to dialysis and subsequent SDS-PAGE and Western-blot analysis to confirm the presence of protein. PR2 and PR3 proteins in the first phase failed to produce. We made new PR3 protein expression. But before expression sequence of the signal (secretory) peptide was removed from the gene of PR3 peptide. After removing the signal sequence of protein expression was successful. The signal peptide probably has a negative effect on the correct protein packing, which then passed into inclusion bodies. The same procedure for removal of the signal sequence is therefore necessary to produce the protein PR2.
