The Effect of Temperature on Shoot Apex Development Rate in Winter Wheat with Diferent Photoperiodic Sensitivity

Abstract:

Tato práce se zabývá problematikou týkající se mechanismů řízení přechodu rostlin pšenice z vegetativní do generativní fáze, a to hlavně z hlediska genů, které reagují na teplotu a délku dne a dalších faktorů, které mohou proces kvetení ovlivnit. V praktické části byl proveden experiment se čtyřmi odrůdami pšenice ozimé, které byly kultivovány za řízených podmínek v klimaboxech a průběžně hodnoceny z hlediska fenologické fáze na základě počtu odnoží a stupně vývoje růstového vrcholu. Bylo zjištěno, že se zvyšující se teplotou se vývoj zrychluje. Současně byly z rostlin odebírány vzorky na následné molekulární analýzy, jejichž cílem bylo stanovení normalizované relativní exprese vybraných genů zapojených ve změně fáze. U genů PPD1, FDL2 a HOX1 nebyly zjištěny žádné zásadní reakce v souvislosti s teplotou, u VRN1 byl indikován nárůst exprese ve vyšší teplotě. U genu VRN3 hladina exprese odrůd citlivých na fotoperiodu výrazně převyšovala expresi naměřenou u odrůd necitlivých.

Abstract:

This thesis deals with the mechanisms, that are involved in the transition from vegetative to reproductive development in wheat, focusing on the regulatory genes, which respond to the temperature, photoperiod and other factors, that may affect the flowering process. In the practical part, an experiment with four varieties of winter wheat was perfomed. Plants were cultivated under controled conditions and continuosly evaluated in the terms of phenological phase based on parameters such as the number of tillers and developmental phase of the apex shoot. Simultaneously, the samples were taken from the plants for subsequent molecular ana-lyses, the aim of which was to determine the normalized relative expression of selected genes involved in the flowering pathway. No significant temperature-related reactions were found for PPD1, FDL2 and HOX1 genes, and an increase in higher temperature was indicated for VRN1. In the VRN3 gene, the expression level of photoperiod-sensitive varieties significantly exceeded the expression in photoperiod-insensitive varieties.
