NMR metabolomics profiling of cancer

Abstract:

This study is aimed on the potential of NMR spectroscopy for the analysis of metabolites (aminoacids, carbohydrates) in the cells of melanoma and glioma. Metabolites are inevitable components of biological systems. The change of their metabolic profiles takes place in a number of diseases (e.g. cancer). This study is focused on the main steps of obtaining NMR spectra of metabolites, preparation and time stability of the samples, storing of the samples and data preprocessing. Several metabolic profiles of melanomas with added inhibitors (Sorafenib, Rotenone, 2-DG) are demonstrated here. A library of aminoacids and several metabolites of Cori and Krebs cycle is also included, where the locations of the signals of individual metabolites are shown and described. Acquired data and knowledge will help in better understanding of the tumor metabolism of melanoma and its individual metabolites in the cellular medium.

Abstract:

Tato práce se zaobírá studiem potenciálu NMR spektroskopie pro analýzu metabolitů (AMK, cukrů) v buňkách melanomu a gliomu. Metabolity jsou nezbytnými komponentami biologických systémů. V řadě onemocnění (např. rakovina) dochází ke změně jejich metabolických profilů. Tato práce se zabývá hlavními kroky v získávání NMR spekter metabolitů, přípravou vzorku a jeho časovou stabilitou, uskladněním vzorku a předzpracováním dat. Je tu demonstrovaných několik metabolických profilů melanomu s přidanými inhibitory (Sorafenib, Rotenon, 2-DG). Taktéž byla vytvořená knihovna aminokyselin a některých metabolitů Coriho a Krebsového cyklu, v které jsou znázorněné a popsané polohy signálů jednotlivých metabolitů. Získaná data a poznatky umožní lépe pochopit nádorový metabolizmus melanomu a jeho jednotlivých metabolitů v buněčném prostředí.
