The determination of metalloproteinases-9 in patients with cardiovascular disease

Abstract:

Vzhledem k rostoucímu počtu pacientů s kardiovaskulárním onemocněním, hledáme nové laboratorní markery, které nám budou co nejlépe popisovat stav pacienta. Mezi tyto nadějné markery patří i matrixová metaloproteinasa 9. Matrixová metaloproteinasa je enzym, účastnící se za fyziologických podmínek remodelace extracelulární matrix, která je nezbytná v procesu vývoje, hojení apod. Špatná regulace její aktivity je ale příčinou mnoha onemocnění, včetně kardiovaskulárního. Tato práce je zaměřena na optimalizaci metody pro stanovení matrixové metaloproteinasy 9 a na stanovení jejích hladin u zdravé populace a pacientů se stabilní anginou pektoris . Těmto pacientům byl následně implantován koronární stent. Metoda pro stanovení matrixové metaloproteinasy 9 byla úspěšně optimalizována. Získaná data ukázala, ţe mezi hladinami u pacientů se stabilní anginou pektoris a zdravou populací není rozdíl. Po implantaci ale významně rostou hladiny celkové metaloproteinasy. Dále je z výsledků patrná korelace s hladinami C-reaktivního proteinu a moţná závislost hladin metaloproteinas 9 na jiţ v minulosti provedené koronární intervenci a druhu pouţitého stentu.

Abstract:

Due to an increasing number of patients suffering from cardiovascular disease we looked for new laboratory markers which would describe the patient's status in the best way. One of the prospective markers was matrix metalloproteinase 9. Matrix metalloproteinase 9 is an enzyme involved in the physiological process of remodelling the extracellular matrix, a necessary part of the process of development, healing, etc. However, poor regulation of its activity may cause many diseases, including cardiovascular disease. This study focuses on the optimization method of measuring matrix metalloproteinase 9 and determining its level in healthy people and patients with persistent angina pectoris. A coronary stent was implanted to these patients. The method of measuring matrix metalloproteinase 9 was successfully optimized. The collected data showed that there was no difference between the levels in patients withpersistent angina pectoris and healthy people. Levels of metalloproteinase significantly increased after implantation. Furthermore, the results suggest a correlation between C-reactive proteins and the possible relation between levels of metalloproteinase-9 and previous subcutaneous coronary intervention types of stent used.
