Interaction of Wnt and FGF signalling

Abstract:

Signalizační kaskády Wnt a FGF, často označované za morfogenetické dráhy, se zásadně podílejí na vývoji jedince již od ranné embryogeneze. Obě dráhy ovlivňují ve velké míře určení, tvar a prostorovou orientaci vyvíjející se tkáně. Přenos signálu probíhající pomocí desítek ligandů, mnoha receptorů a koreceptorů, se v cytoplazmé šíří pomocí složité sítě kináz a jejich substrátů. Hlavním cílem signalizace je řízená transkripce genů, jejichž exprese konroluje buněčnou proliferaci a diferenciaci. Poruchy obou signalizací vedou často k embryonálně letálním defektům, bodové mutace receptoru FGFR-3 vedou ke vzniku achondroplazie a nadbytečná aktivita signalizace Wnt byla popsána u většiny nádorových onemocnění. Při vývoji končetiny jsou mezenchymální buňky vytvářející základy budoucí kosti vystaveny signálům FGF z apikální ektodermální lišty (AER) podporujícím jejich dlouživý růst a nediferencovaný charakter. Zároveň jsou tytéž mezenchymální buňky vystaveny signálům Wnt z tenké vrstvičky ektodermálních buněk tvořících základ budoucí epidermis. Signalizace Wnt působící dorso-ventrálně brání diferenciaci chondrocytů a podporuje proliferaci mezenchymálních buněk. Spolupůsobení obou drah na genovou expresi chondrocytů a jejich mezenchymálních předchůdců bylo popsáno teprve nedávno a molekulární mechanismy, kterými k tomu dochází nebyly dosud popsány vůbec. Ve své práci nejprve shrnuji dosavadní poznatky týkající se signalizací Wnt a FGF a jejich interakce při chondrogenezi. Výsledky v experimentální části ukazují, že spolupůsobením ligandů FGF2 a Wnt3a dochází k markantnímu navýšení exprese promotoru, který je pod kontrolou Wnt dráhy. Zesílení signalizace je s největší pravděpodobností způsobeno zvýšenou fosforylací Lrp5/6, tedy koreceptoru Wnt, kinázou Erk1/2, která odpovídá na stimul FGF. Další experimenty naznačují, že obdobný efekt na expresi cílových genů signalizace Wnt mají i mutace receptoru FGFR-3 zodpovědné za vznik thanatoforické dysplazie I a II typu a hypochondroplazie.

Abstract:

The Wnt and FGF signaling cascades are morphogenetic pathways important in development of an individual from very early embryogenesis. Both pathways affect cell fate, shape and space orientation of growing tissues. Signal can be transduced by tens of ligands and many receptors and co-receptors into the cytoplasm where it is spread and modulated by a number of kinases and other interacting proteins. Main effects of signaling are proliferation and differentiaiton of target cells achieved by controled gene expression. Disorders in these pathways result in embryonaly lethal fenotypes and even point mutation in the receptor FGFR-3 may cause skeletal disorders like achondroplasias. Overactive Wnt signaling in adult organism can be found in majority of cancers. Mesenchymal cells are stimulated by FGF signal from apical ectodermal ridge (AER) to proliferate and remain undifferentiated in order to elongate the forming limb during limb development. The same cells are in the same time stimulated by Wnt signal from a thin ectodermal cell layer of a forming epidermis. Dorso-ventraly active Wnt signal is also preventing differentiation of mesenchymal cells into chondrocytes and supports their proliferation. Synergistic effect of both pathways on gene expression of mesenchymal cells and chondrocytes has been discovered recently and molecular mechanisms mediating these events were not described at all. Theoretical part of this work is focused on summarizing the current knowledge about Wnt and FGF pathways and their role in chondrogenesis. In vitro results in the experimental part of my work are bringing strong evidence about synergistic positive effects of FGF2 ligand on the Wnt driven transcription. Such increase in the signaling response is mediated by phosphorylation of the Wnt pathway co-receptor Lrp5/6, mediated by FGF-dependent activity of Erk1/2 kinase. Further experiments show that similar effects on the Wnt-driven transcription are obtained when cells activate FGF pathway by mutated forms of receptor FGFR-3 responsible for hypochodroplasia and thanatophoric dysplasia I and II.
