Determination of kinetic parameters of enzymatic hydrolyses of acetylcholine and acetylthiocholine

Abstract:

Práce byla zaměřena na sledování kinetiky neinhibovaných a inhibovaných enzymatických hydrolýz acetylcholinu (ACh) a acetylthiocholinu (ATCh) pomocí acetylcholinesterázy (AChE) a butyrylcholinesterázy (BChE). K inhibici hydrolýz prováděných in vitro bylo použito 7 inhibitorů s imidazoliofanovou strukturou. Z výsledků práce vyplývá, že studované látky vykazují dobrou anticholinesterázovou aktivitu. Ve většině případů lze inhibitory popsat modelem kompetitivní nevratné inhibice. Požadavek kladený na cholinesterázové inhibitory (ChEI) (centrální účinnost) však nesplňují a tudíž by se neosvědčily jako inhibitory mozkové AChE.

Abstract:

This thesis was focused on monitoring of the kinetics of enzymatic hydrolyses of acetylcholine (ACh) and acetylthiocholine (ATCh) by acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). The inhibited hydrolyses were carried out in vitro with 7 inhibitors with imidazoliophane structure. From obtained results it is possible to conclude that tested compounds show sufficient anticholinesterase activity. The course of most of inhibited hydrolyses could be described by model of competitive irreversible inhibition. However tested compounds did not fill one of the basic requirements required on cholinesterase inhibitors (i.e. central effect) and that is why they would not be suitable inhibitors of brain acetylcholinesterase.
