Genetic variation and clinical heterogeneity in Cystic Fibrosis

Abstract:

Cystická fibróza (CF) je nejčastějším autozomálně recesivně dědičným onemocněním u kavkazské populace. Primární příčinou tohoto onemocnění jsou mutace v genu CFTR, který kóduje protein fungující jako chloridový kanál. Odlišnosti v průběhu onemocnění u jedinců se stejnými mutacemi tohoto genu jsou způsobeny, kromě dalšího, také sekvenčními variantami CFTR genu mimo kauzální mutace. Samy o sobě tyto varianty narušují funkci CFTR proteinu pouze malou měrou a jejich přítomnost tak nevede k rozvoji klasické formy CF, avšak mohou modulovat účinek kauzálních mutací. V přítomnosti více takovýchto variant na jednom chromozomu se navíc mohou jejich účinky na funkci proteinu kumulovat. Cílem této práce bylo zavedení analýzy variant IVS8TG(m)T(n) a M470V, u nichž je předpokládán modifikující vliv na průběh onemocnění. K tomu byla využita metoda analýzy křivek tání, alelově-specifické PCR a kvantitativní PCR.

Abstract:

Cystic fibrosis is one of the most common autosomal recessive genetic disorders of the Caucasian population. The primary cause of the disease are mutations of the CFTR gene, that encodes the CFTR protein, which fulfills the function of chloride channel. Differences in the course of this disease in individuals with the same mutations of the CFTR gene are caused, among other, by sequence variations of the CFTR gene. By themselves, these variations affect the function of the CFTR gene to only a small extent and their presence does not cause classic cystic fibrosis, however they could modulate the signs of "main" mutations and if more of the variations are present on one chromosome, their effects on the protein function could add up. The goal of this thesis is to implement an analysis of the variations IVS8TG(m)T(n) and MV470V, which are expected to have a modifying influence on the course of the disease. Melting curve analysis, multiplex allele specific fluorescent PCR and quantitative PCR were used.
