Use of intraperitoneal application of ganoderma in the rat

Abstract:

Původem čínská houba Ganoderma lucidum je komerčně dostupný doplněk stravy podporující imunitní systém člověka. Je velice rozšířenou a hojně užívanou houbou po celém světě. Cílem této práce bylo odhalit vliv této houby Ganoderma lucidum na imunitní buňky a zda-li vyvolá zánětlivou reakci. Vhodným prostředím pro kontakt s imunitními buňkami je právě intraperitoneální dutina potkana. Tedy potkanům kmenu Wistar bylo aplikováno určité množství koncentrované houby a po 24 h byla provedena intraperitoneální laváž. Ze získaného vzorku byly stanoveny absolutní počty imunitních buněk a jejich relativní podíl po 24 hodinách od aplikace, a to různými metodami. Byly využity měřící přístroje jako cell counter, průtokový cytometr a světelný mikroskop. Získané výsledky byly srovnávány s kontrolní skupinou potkanů, kterým bylo aplikováno PBS tedy fyziologický roztok. Porovnání výsledků Ganodermy se značně lišily od výsledků kontroly. Absolutní počty u Ganodermy byly zvýšené. Diferenciální počty byly u Ganodermy také zvýšeny a vyšly statisticky významné. Konkrétně u Ganodermy došlo k zvýšení neutrofilů a makrofágů. Došlo také k poklesu histiocytů. Podíl lymfocytů nevyšel statisticky významný. Po aplikaci Ganodermy tedy došlo k migraci neutrofilů a zvýšení počtu makrofágů v peritoneální dutině. Došlo k potenciální zánětlivé reakci a podpoře imunitního systému.

Abstract:

The origin of the Chinese fungus Ganoderma lucidum is a commercially available food supplement that supports the human immune system. It is a widespread and widely used fungus around the world. The aim of this work was to reveal the effect of this fungus Ganoderma lucidum on immune cells and whether it causes an inflammatory response. The rat's intraperitoneal cavity is a suitable environment for contact with immune cells. Thus, a number of concentrated fungi were administered to Wistar rats and intraperitoneal lavage was performed after 24 hours. From the obtained sample, the absolute numbers of immune cells and their relative proportion were determined 24 hours after application, by various methods. Measuring instruments such as a cell counter, a flow cytometer and a light microscope were used. The results obtained were compared with a control rat treated with PBS, i.e. saline. Comparison of Ganoderma results differs significantly from control results. Absolute counts in Ganoderma have been improved. Differential numbers were also increased and significant statistics for Ganoderma. In Ganoderma in particular, neutrophils and macrophages increased. There was also a decrease in histiocytes. The proportion of lymphocytes was not statistically significant. Thus, after administration of Ganoderma, neutrophils migrated and the number of macrophages in the peritoneal cavity increased. There was a potential inflammatory response and support for the immune system.
