Enzyme kinetics in teaching analytical chemistry

Abstract:

This Bachelor’s thesis deals with enzyme kinetics, examining the time-course of the reaction catalyzed by the enzyme horseradish peroxidase from the group of oxidoreductases. The enzyme activity was measured spectrophotometrically and fluorimetrically. The theoretical part of my thesis is devoted to the issue of enzymes, contains a more detailed description of the kinetics of enzymatic reactions with one substrate and at the end of this section describes the basic principles of optical methods and focusing on their use in enzyme analysis. The practical part is mainly engaged in measuring enzyme kinetics of the reaction system containing the enzyme horseradish peroxidase, substrate Amplex UltraRed with constant concentration and hydrogen peroxide solution of different concentrations. Part of my thesis is the study of inhibition of the enzyme reaction, too. The result of this thesis is calculated initial speed v0 and rate constants k2 of the enzyme reaction at a constant concentration of the substrate in the reaction system and various concentrations of hydrogen peroxide, as well as experiment instruction of dealing with the enzyme kinetics for teaching analytical chemistry.

Abstract:

Bakalářská práce se zabývá enzymovou kinetikou, zkoumáním časového průběhu reakce katalyzované enzymem křenovou peroxidázou ze skupiny oxidoreduktáz. Enzymová aktivita byla měřená spektrofotometricky a fluorimetricky. Teoretická část je věnovaná problematice enzymů, obsahuje podrobnější popis kinetiky enzymové reakce s jedním substrátem a v závěru této části jsou popsané základní principy použitých optických metod i se zaměřením na jejich využití v enzymové analýze. Praktická část se především zabývá měřením enzymové kinetiky reakčního systému obsahujícího enzym křenovou peroxidázu, substrát Amplex UltraRed s konstantní koncentrací a roztok peroxidu vodíku o různých koncentracích. Součástí praktické části je i studium inhibice enzymové reakce. Výsledkem této práce jsou vypočítané počáteční rychlosti v0 a rychlostní konstanty k2 enzymové reakce s konstantní koncentrací substrátu v reakčním systému a různými koncentracemi peroxidu vodíku a taktéž zpracovaný návod k úloze zabývající se enzymovou kinetikou určený do praktika z analytické chemie.
