Infection of poultry with less frequent Salmonella enterica serovars

Abstract:

Drůbež je pro člověka častým zdrojem netyfoidních salmonel. Proto se přistupuje k opatřením, která mají za úkol snížit prevalenci salmonel u kuřat a potažmo snížit incidenci humánních salmonelóz. Jedním z možných opatření je použití probiotik nebo produktů kompetitivní exkluze. Dalším stupněm ochrany kuřat proti infekci je vakcinace. V první části práce jsme se proto zabývali srovnáním imunitní odpovědi vylíhlých kuřat na infekci různými kmeny S. Enteritidis, Typhimurium a Infantis a funkcí O-antigenu při této interakci. Nejdříve byla vylíhlá kuřata infikována sedmi různými divokými kmeny S. Enteritidis, Typhimurium a Infantis a jejich rfaL mutanty a následně byla určena imunitní reakce kuřat na infekci. Všechny divoké kmeny vyvolaly větší zánětlivou odpověď než kterýkoli rfaL mutant. Imunitní odpověď kuřat na infekci salmonelami byla závislá na přítomnosti O-antigenu a značně se lišila bez ohledu na příslušnost k sérovaru. V další části práce jsme zjišťovali, zda mikroflóra získaná od různě starých donorových slepic ochrání kuřata stejnou měrou proti infekci S. Enteritidis 147. Vylíhlá kuřata byla proto kolonizována směsí bakterií ze slepého střeva 1, 3, 16, 28 nebo 42týdenních donorů. Po sedmi dnech byla kuřata infikována S. Enteritidis 147. Dále byl sledován profylaktický či terapeutický účinek mikroflóry získané od 35týdenních donorů na infekci S. Enteritidis 147. Jak kultivace salmonel, tak exprese genů ve slepém střevě kuřat potvrdily, že mikroflóra získaná od 3týdenních a starších slepic chránila kuřata proti infekci S. Enteritidis 147. Mikroflóra získaná od 35týdenních slepic byla schopna ochránit kuřata již 24 h po kolonizaci. V poslední části práce jsme testovali účinnost vakcíny sestávající z oslabených kmenů S. Enteritidis 147, Typhimurium 2002 a Infantis 1516 proti infekci divokými kmeny sérovarů obsažených ve vakcíně a proti infekci S. Agona, Dublin a Hadar FA2033 (heterologní sérovary). Také byl zjišťován účinek střevní mikroflóry podané současně s vakcínou. Kuřata byla první den života vakcinována a po 20 dnech byla infikována homologními a heterologními sérovary. Ochranný účinek vakcíny byl určen na základě počtu salmonel v játrech pokusných kuřat. Pokusy prokázaly účinnost připravené vakcíny proti divokým kmenům sérovarů obsažených ve vakcíně a proti izolátům naležícím k sérovaru Dublin a Hadar FA2033. Mikroflóra podaná současně při vakcinaci chránila kuřata ještě před indukcí specifické imunity.

Abstract:

Poultry is common source of non-typhoid Salmonella for humans. Therefore different interventions are designed to reduce the prevalence of Salmonella in chickens and thereby the incidence of human salmonellosis is decreasing. One such intervention is the use of probiotic, competitive exclusion products or vaccination. In the first part of the dissertation, we were interested in the comparison of chicken immune response to infection with different strains of S. Enteritidis, Typhimurium and Infantis, and a role of O-antigen in these interactions. Newly hatched chickens were infected with seven wild type strains of S. Enteritidis, Typhimurium and Infantis as well as their rfaL mutants. All wild type strains stimulated greater inflammatory response than any of the rfaL mutants. Chicken immune response to Salmonella infection was found to be dependent on the presence of O-antigen and greatly differed regardless serovar classification. In the next part of the dissertation, we tested whether microbiota from donor hens of different age will equally protect chickens against S. Enteritidis 147 infection. Newly hatched chickens were inoculated with cecal extracts from 1-, 3-, 16-, 28-, and 42-week-old donors, and 7 days later chickens were infected with S. Enteritidis 147. Furthermore, we tested the prophylactic or therapeutic effect of microbiota obtained from 35-week-old donors to infection with S. Enteritidis 147. Microbiota obtained from 3-week-old or older donors protected newly hatched chickens against S. Enteritidis 147 challenge. Microbiota from 35-week-old hens protected chickens even 24 h after administration. In the last part of the dissertation, we tested the efficacy of a vaccine consisting of attenuated strains of S. Enteritidis 147, Typhimurium 2002 and Infantis 1516 against the challenge with the same serovars and with S. Agona, Dublin and Hadar FA2033. The protective potential of intestinal microbiota administered simultaneously with the vaccine was also investigated. Chickens were vaccinated on the first day of life and after 20 days were challenged with homologous and heterologous serovars. The protective effect was determinated by bacterial enumeration in liver of experimental chickens. We showed that the vaccine consisting of attenuated Salmonella strains protected chickens against challenge with the wild type strains of the same serovars and may protect also against isolates belonging to other serovars. Co-administration of cecal microbiota and vaccine protects chickens before induction of specific immunity.
