The role of salicylic acid in the defence response of tomato plants

Abstract:

Kyselina salicylová (SA) má zásadní roli v obranných reakcích rostlin. Rozpoznání SA jejími receptory vede k expresi genů závislých na SA. V důsledku této exprese je SA dále biosyntetizována i metabolizována a jsou syntetizovány systémové signály, které pak spolu s SA vedou k systémově navozené rezistenci. Ukazuje se však, že tyto procesy se mohou lišit u jednotlivých rostlin. Cílem této práce bylo porovnat úlohu SA v obranných reakcích rostlin řádu Solanaceae, konktrétně Nicotiana tabacum a Solanum lycopersicum. Výsledky měření ukázaly, že se metabolismus SA v tabáku a v rajčeti liší. V tabáku se SA pomalu metabolizuje na její glukosid, zatímco v rajčeti SA spouští svoji biosyntézu a akumuluje se jako volná SA. Dále bylo zjištěno, že SA v tabáku a rajčeti různě ovlivňuje expresi PR genů. V tabáku se vlivem SA zvýšila exprese PR1 a PR5 genů, avšak nikoliv PR3 genu, zatímco v rajčeti se vlivem SA zvýšila exprese CHIT4 genu, avšak nikoliv P6 a HEV2 genů. Také bylo potvrzeno, že předběžné ošetření SA indukovalo rezistenci vůči Pseudomonas syringae tabaci v tabáku, zatímco u rajčete nemělo žádný významný efekt na rezistenci vůči P. syringae DC3000. Výše uvedená zjištění přispívají k výzkumu obranných reakcí rajčete.

Abstract:

Salicylic acid (SA) plays an essential role in the defense responses of plants. Perception of SA by its receptors leads to the expression of SA-dependent genes, which in turn promotes the biosynthesis of SA and its metabolism and biosynthesis of systemic signals that, together with SA, lead to systemic acquired resistance. However, it appears that these effects may vary between different plants. This work aimed to compare the activation of immunity responses by SA between plants from the Solanaceae family, specifically between Nicotiana tabacum and Solanum lycopersicum. Firstly, it was demonstrated that the metabolism of SA differs in tobacco and tomato plants. The findings suggested that SA is slowly metabolized to a SA glucoside in tobacco. In contrast, in tomato plants, SA appears to enhance its biosynthesis and accumulate as free SA. Secondly, it was demonstrated that in tobacco and tomato plants, SA influences the expression of PR genes differently. In tobacco, SA increased the expression of PR1 and PR5 genes but not PR3 gene, whereas in tomato, it increased the expression of CHIT4 gene but not P6 and HEV2 genes. Finally, it was demonstrated that pretreatment with SA induced resistance to Pseudomonas syringae tabaci in tobacco but had no significant effect on the resistance of tomato to P. syringae DC3000. These findings contribute to the research on the defense mechanisms of tomato plants.
